FOXC2 和E-cadherin 在宫颈鳞癌组织中的表达及临床意义

摘要
目的:通过检测叉头框蛋白C2(forkhead box C2,FOXC2)和E-钙黏蛋白(E-cadherin,E-cad)在宫颈鳞癌组织中的表达,探讨二者在宫颈鳞癌发生发展中的作用。方法:采用免疫组织化学SP法检测FOXC2,E-cad在40例正常宫颈组织,65例宫颈上皮内瘤变(cervical intraepithelial neoplasias,CIN)组织和98例宫颈鳞癌组织中的表达;分析FOXC2,E-cad表达与宫颈鳞癌临床病理因素的关系;用Spearman秩相关分析FOXC2和E-cad在宫颈鳞癌组织中表达的相关性。结果:FOXC2,E-cad在宫颈鳞癌组、CINII~III组、CINI组和对照组四种组织中的阳性表达率和染色强度差异有统计学意义(χ2=49.801,35.512,P<0.05;F=15.278,18.375,P<0.05);FOXC2和E-cad在不同FIGO分期、浸润深度和有无淋巴结转移的宫颈鳞癌组织中阳性表达率差异有统计学意义( χ2=3.752~9.362, P<0.05);FOXC2,E-cad在宫颈鳞癌组织中表达呈负相关(r=−0.658,P<0.05)。结论:FOXC2在宫颈鳞癌中呈高表达,E-cad呈低表达,且两者表达呈负相关,两者在宫颈鳞癌发生发展过程中可能存在相互作用。

Abstract
Objective: To detect the expression of forkhead box C2 (FOXC2) and E-cadherin (E-cad) in cervical squamous cancer tissues, and to explore the role of FOXC2 and E-cad in the occurrence, invasion and metastasis of cervical cancer. Methods: Immunohistochemical SP method was used to detect the expression of FOXC2 and E-cad in 40 cases of normal cervical tissues, 65 cases of cervical intraepithelial neoplasias (CIN) tissues, and 78 cases of cervical cancer squamous tissues. The relationships between the two proteins expression and clinical pathological characteristics of cervical cancer were analyzed. Spearman rank correlation was used to analyze the correlation of the expression of FOXC2 and E-cad in cervical cancer tissues. Results: The positive expression rate and staining intensity of FOXC2 and E-cad in four tissues of cervical squamous cell carcinoma, CIN II–III group, CIN I group and control group were statistically significant (χ2=49.801, 35.512, P<0.05; F=15.278, 18.375, P<0.05). The expression of FOXC2 and E-cad in cervical carcinoma was related to FIGO stage, depth of invasion and lymph node metastasis (χ2=3.752–9.362, P<0.05). The expressions of FOXC2 and E-cad were negatively correlated in cervical cancer tissues (r=−0.658, P<0.05). Conclusion: FOXC2 shows high expression in cervical squamous cancer tissues, while E-cad is low expressed. Meanwhile, a negative correlation is observed between the two proteins. They may interact in the course of carcinogenesis and development of cervical squamous cancer.
