PD-1及其配体在原发性干燥综合征患者唇腺中的表达及临床意义

摘要
目的:观察干燥综合征(Sjogren’s syndrome,SS)患者唇腺活检组织中程序性死亡分子-1(programmed death-1,PD-1)、程序性死亡配体-1(programmed death ligand-1,PD-L1)、程序性死亡配体-2(programmed death ligand-2,PD-L2)的表达,以及其与临床病理特征的相关性。方法:对所有患者和正常对照组的唇腺活检标本进行HE染色观察淋巴细胞浸润程度,免疫组织化学观察PD-1,PD-L1和PD-L2的表达。结果:SS患者唇腺活检组织中PD-1,PD-L1和PD-L2的表达较正常对照组明显增强。SS患者唇腺活检标本淋巴细胞的浸润程度与PD-1和PD-L2的表达强度无明显相关性,与PD-L1的表达强度呈正相关。SS患者疾病活动度评分与PD-1,PD-L1及PD-L2的表达强度均无明显相关性。SS患者疾病损害评分与PD-1的表达强度无明显相关性,与PD-L1和PD-L2的表达强度呈负相关。结论:PD-1,PD-L1及PD-L2在SS患者唇腺活检组织中的表达明显增强,PD-1/PD-L信号通路可能在SS的免疫病理损害中起重要作用。

Expression and clinical significance of PD-1 and its ligands in salivary glands of patients with Sjogren’s syndrome

Abstract
Objective: To investigate the expression and its relativity with the clinicopathological characteristics of programmed death-1 (PD-1) and its ligands PD-L1 and PD-L2 in salivary glands of patients with Sjogren’s syndrome (SS). Methods: We used HE staining to observe the lymphocyte infiltration while used immunohistochemistry to observe the expression of PD-1, PD-L1 and PD-L2 in both patients’ and normal-controlled group’s salivary glands biopsies. Results: The expression of PD-1, PD-L1 and PD-L2 in salivary glands biopsies of patients with SS significantly enhanced compared with the normal-controlled group. There were no significant relevance between the lymphocyte infiltration and the expression degrees of PD-1 and PD-L2 in salivary glands biopsies of patients with SS. However, lymphocyte infiltration had positive correlation with the expression degrees of PD-L1 in salivary glands biopsies of patients with SS. There were no significant relevance between the disease activity index of patients with SS and the expression degree of PD-1, PD-L1 and PD-L2. There was no significant relevance between the disease damage index of patients with SS and the expression degree of PD-1, while the disease damage index was negatively correlated with the expression degree of PD-L1 and PD-L2. Conclusion: The expression of PD-1, PD-L1 and PD-L2 significantly enhance in salivary glands biopsies of patients with SS. Therefore, PD-1/PD-L pathway may play a big role in immune pathological damage of SS.
