细胞内脂滴及其相关Rab蛋白的研究进展

摘要
脂滴作为细胞内储存中性脂的主要部位,普遍存在于各类物种的各种细胞中。脂滴以中性脂肪为核心,表面覆盖单层磷脂结构,并镶嵌多种蛋白质。近年来的蛋白组学研究表明,脂滴相关蛋白通过静电或化学结合等方式定位在脂滴的表面,并可能参与调控脂代谢、膜转运以及信号转导等多个生理过程,进而揭示了脂滴在细胞内并不是一个简单的能量储存器,而是一个活跃的多功能细胞器。然而,Rab家族蛋白作为脂滴蛋白的一种,在介导脂滴与其它细胞器之间的信息交流可能发挥重要作用,目前,对其作用的具体机制仍未完全清楚。本文就近年来对脂滴的生物学研究及其相关的Rab蛋白研究作一综述。

Abstract
Lipid droplet is an intracellular structure that consists of a neutral lipid core surrounded by a monolayer of phospholipids. Recently, more and more functional proteins have also been found to be associated with lipid droplet by proteomic studies. These proteins mosaic on the surface of lipid droplet via electrostatic or chemical way, and they may mediate lipid metabolism, membrane traffic, protein degradation, and signal transduction. Therefore, lipid droplet not only store lipids for catabolism but also function as an dynamic activities organelle for cellular lipid anabolism. Rab family proteins have been identified as major proteins on the surface of lipid droplet. Recent works demonstrate that rab family proteins may play an important role in information exchange between lipid droplet and other organelles, but the mechanism of its action has not yet entirely clear. In this paper, recent research on the biology and related research lipid droplet rab proteins are reviewed.
