miR-136通过抑制PPP2R2A表达调控胃癌细胞SGC7901凋亡

摘要
目的:检测miR-136对胃癌细胞SGC7901增殖和凋亡的影响及其机制。方法:分别利用MTT实验和Annexin V-PI实验检测转染miR-136 inhibitor的胃癌细胞SGC7901增殖和凋亡。利用western blot和双荧光素酶报告基因技术检测miR-136的靶基因。结果:抑制miR-136表达可以明显抑制SGC7901细胞增殖和诱导其凋亡。miR-136能够与PPP2R2A mRNA 3'-UTR区结合。结论:抑制miR-136表达能够促进PPP2R2A蛋白表达,进而引起胃癌细胞凋亡。

Abstract
Objective: To detect the effects and mechanisms of miR-136 on gastric cancer cell SGC7901. Methods: The proliferation and the apoptosis of SGC7901 cells after transfected with miR-136 inhibitor were detected by MTT assay and Annexin V-PI assay, respectively. Western blot and dual luciferase reporter gene were used to detect the target gene of miR-136. Results: Inhibition of miR-136 could inhibit the proliferation and induce apoptosis in SGC7901 cells. miR-136 could combine with 3'-UTR of PPP2R2A mRNA. Conclusion: Inhibition of miR-136 could induce PPP2R2A expression and apoptosis in SGC7901 cells.
