翻译后组蛋白乙酰化与骨代谢的研究进展

摘要
表观遗传学是指研究基因的核苷酸序列不发生改变的情况下,基因表达的可遗传的变化的一门遗传学分支学科,主要包括miRNA介导的转录后调控、翻译后的组蛋白修饰以及DNA甲基化。其中,组蛋白乙酰化作为组蛋白修饰中研究最广的内容,已经被证实能影响多种生理以及病理过程。在骨代谢中,组蛋白乙酰化影响着成骨细胞和破骨细胞的分化成熟,在骨形成和骨吸收动态平衡中起着至关重要的作用。本文综述了组蛋白乙酰化对骨代谢的影响,以及各种组蛋白乙酰化酶(histone deacetylase,HATs)和组蛋白去乙酰化酶(histone deacetylase,HDACs)在其中的作用,希望为下一步探讨组蛋白乙酰化对骨代谢相关疾病的影响提供方向。

Abstract
Epigenetics is the study of mechanisms able to influence gene expression in heritable manner without altering the DNA sequence. These mechanisms include miRNA-mediated post-transcriptional regulation, post-translational histone modifications and DNA methylation. Among all the histone modification, histone acetylation is reserched most widely. And it has been confirmed to influence many physiological and pathological progresses. In bone metabolism, histone acetylation influences osteoblastogenesis and osteoclastogenesis. Thus, it plays an important role in bone homeostasis. This review introduces the roles of histone acetylation, HDACs and HATs in bone metabolism, and we hope it will provide a new direction for the research of bone-metabolism related diseases.
