摘要

BCL2和腺病毒E1B19kDa相互作用蛋白3(BNIP3)与BCL2家族蛋白具有同源性,都含有BH3结构
域,是BH3-only亚家族的非典型成员之一。高表达的BNIP3绝大多数定位在线粒体外膜。虽然机
体存在许多其他可以诱导线粒体自噬及凋亡的蛋白,效果甚至更优于BNIP3,但是BNIP3将诱导线
粒体自噬和凋亡联系起来,并且这两种功能之间的转化与细胞最终命运关系密切,这使得它比其
他单独促凋亡或促自噬的蛋白更具有研究意义。本文总结BNIP3诱导细胞线粒体自噬和凋亡对肿
瘤的影响,并探讨其发挥功能的可能机制。

Abstract

BCL2 and adenovirus E1B19 kDa interacting protein 3 (BNIP3) is protein with homologous to BCL2 in the BH3 domain, which are the atypical member of the BH3-only subfamily. When BNIP3 over-expressed, the vast majority of them are located in the mitochondrial outer membrane. Although many other proteins may induce cell mitophagy or apoptosis more efficiently than BNIP3, the ability of BNIP3 associating mitophagy with apoptosis and ultimately affecting the fates of cell makes it more significance than other single promote mitophagy or apoptosis protein. Here we review the role of BNIP3 in mitophagy and apoptosis of the tumor, and discuss potential mechanisms by which they may function.
