关于透射电镜样品污染问题的探讨

摘要

在透射电镜样品制备过程中,经常会遇到超薄切片被污染的问题。本文分析了造成超薄切片污染 的原因,并针对超薄切片制作过程中的污染,提出了改善切片室环境、铜网支持膜、合理使用切 片刀及刀槽的改进措施,针对切片电子染色过程中的污染,提出了在染液配制过程中严格遵守操 作程序、在染色操作过程中严格坚持正确的方法等注意事项,并阐明了必要的防范措施,这些将有 效避免电镜样品污染现象。

Abstract

In the process of tem sample preparation, ultra-thin slicing polluted problems are often occurred. This paper analyzes the causes of ultrathin section pollution and puts forward measures to improve section room environment, copper wire mesh support membrane and the rational use of slicing knife and knife trough to against the pollution in the process of ultrathin section production. Aimed at the pollution in section electronic dyeing process, it also puts forward some operational procedures which are to be strictly complied with in the process of dyeing liquid dispensing, correct methods and matters needing attention in the process of dyeing operations and clarified the necessary measures to effectively prevent electron microscope samples from pollution.
