摘要

目的:探究阿司匹林在调控子宫内膜容受性分子HOXA10表达及胚胎黏附的作用及机制。方法:不同浓度的阿司匹林刺激Ishikawa细胞,相应时间点收取细胞总RNA和蛋白;特异性干扰HOXA10表达的siRNA转染入Ishikawa细胞。分别利用Real-time PCR和Western blot检测HOXA10的mRNA和蛋白水平。利用BeWo细胞球黏附于Ishikawa细胞的模型检测阿司匹林在体外对胚胎黏附的影响。结果:经不同浓度的阿司匹林刺激后,相比于DMSO对照组,Ishikawa细胞中HOXA10的mRNA和蛋白水平都升高,在10−5 mol/L的阿司匹林处理组升高的最明显(P<0.01)。同时,阿司匹林促进BeWo细胞球黏附于Ishikawa细胞上,然而当特异性siRNA干扰HOXA10表达后,阿司匹林对胚胎黏附的促进作用被抑制。结论:阿司匹林可能通过上调子宫内膜容受性分子HOXA10的表达促进胚胎的黏附。

Aspirin promotes embryo implantation in vitro through up-regulation of HOXA10 in Ishikawa cells

Abstract

Objective: To explore the role and mechanism of aspirin in regulating the expression of endometrial receptivity marker gene HOXA10 and embryo implantation. Methods: In this study, Ishikawa cells were treated with different doses of aspirin for indicated time and transfected with siRNAs against HOXA10. The expression of HOXA10 mRNA and protein in Ishikawa cells were determined by Real-time PCR and Western blot. Furthermore, the effect of aspirin on embryo implantation in vitro was determined by an in vitro model of BeWo-Ishikawa cell interaction. Results: Compared with the control group treated with DMSO, the expression of HOXA10 mRNA and protein were up-regulated in Ishikawa cells treated with different doses of aspirin. And, the mRNA and protein levels of HOXA10 were highest in Ishikawa cells treated with aspirin at the dose of 10−5 mol/L (P<0.01). Furthermore, aspirin promoted BeWo spheroids attachment to Ishikawa cells, however, the effect of aspirin on BeWo spheroids attachment was inhibited by HOXA10-specific siRNAs in Ishikawa cells. Conclusion: Aspirin may promote embryo implantation through up-regulation of endometrial receptivity marker gene HOXA10.
