BC-1215抑制三硝基苯磺酸诱导的小鼠肠黏膜炎症反应

摘要
目的:探索F-box蛋白3(F-box protein 3,FBXO3)抑制剂BC-1215对三硝基苯磺酸(trinitrobenzene sulfonic acid,TNBS)诱导的小鼠肠黏膜炎症反应的作用。方法:将Balb/c小鼠随机分为4组,每组10只,2组给予TNBS灌肠诱导结肠炎(实验组),其中1组给予BC-1215腹腔注射,另1组给予磷酸缓冲盐溶液(PBS)腹腔注射;另外2组给予等体积50%乙醇灌肠作为阴性对照组,亦分别给予BC-1215或PBS腹腔注射。5 d后收集结肠组织,通过对比体重、结肠缩短程度、结肠组织苏木精-伊红染色(hematoxylin-eosin staining,HE)及髓过氧化物酶活性评估BC-1215对肠黏膜炎症影响,使用荧光定量PCR(qRT-PCR)检测肠黏膜组织中炎症细胞因子IL-6,TNF-α,IL-1β,IL-23及Th1细胞和Th17细胞相关细胞因子(IFN-γ和IL-17A)和转录因子(T-bet和RORγt)表达。结果:BC-1215抑制TNBS诱导的小鼠体重下降、减轻小鼠结肠缩短程度和炎症细胞浸润,并且抑制小鼠结肠组织中炎症细胞因子表达。结论:BC-1215对TNBS诱导的小鼠结肠炎具有保护作用,对结肠炎具有潜在治疗作用。

Abstract
Objective: To explore the effect of BC-1215 on TNBS-induced colitis in mice. Methods: Balb/c mice were randomly divided into four groups. Two groups were given TNBS enema to induce colitis, and then were intraperitoneally injected with BC-1215 and PBS, respectively. The other two groups were given an equal volume of 50% ethanol enema, and were also injected with BC-1215 or PBS intraperitoneally as a negative control group. Colon tissues were collected 5 days later, HE staining was used to detect pathological damage, and qRT-PCR was used to detect the expression of pro-inflammatory cytokines (IL-6, TNF-α, IL-1β and IL-23), Th1 cells expressed IFN-γ and T-bet, and Th17 cells expressed IL-17A and RORgγt. Results: BC-1215 can significantly alleviate the severity of TNBS-induced colitis, characterized by less body weight loss, moderate colon shortening, slight histological damage and low lower pro-inflammatory cytokines expression in mice. Conclusion: BC-1215 has protective effects on TNBS-induced colitis in mice and has potential therapeutic effects on colitis.
