摘要

目的:探索骨肉瘤患者肿瘤组织和血清中环状RNA hsa_circ_0005721表达水平及临床意义。方法:采用qRT-PCR检测50例骨肉瘤患者癌组织和癌旁组织中hsa_circ_0005721表达水平,探讨hsa_circ_0005721表达水平与患者临床病理参数的关系,Kaplan-Meier生存分析并比较hsa_circ_0005721高低表达患者无瘤生存率及总生存率差异。qRT-PCR法检测30例骨肉瘤、30例骨良性肿瘤和30例健康对照者血清中hsa_circ_0005721相对表达水平,受试者工作特征(receiver operating characters,ROC)曲线分析血清hsa_circ_0005721对骨肉瘤患者的诊断价值。结果:骨肉瘤组织中hsa_circ_0005721表达水平显著高于癌旁组织(P<0.01)。hsa_circ_0005721表达水平与患者Enneking分期和肺转移显著相关(P<0.01);hsa_circ_0005721高表达患者无瘤生存率(P<0.001)和总生存率(P<0.001)均显著低于hsa_circ_0005721低表达患者。hsa_circ_0005721表达水平是影响骨肉瘤患者无瘤生存率(P=0.009)和总生存率(P=0.021)的独立危险因素。骨肉瘤患者血清中hsa_circ_0005721相对表达水平显著高于良性骨肿瘤患者和健康对照者(P<0.001);骨肉瘤患者血清中hsa_circ_0005721区分骨肉瘤和健康对照者的ROCAUC为0.821(95%CI 0.651~0.891),区分骨肉瘤和良性骨肿瘤的ROCAUC为0.701(95%CI 0.601~0.801)。结论:Hsa_circ_0005721在骨肉瘤组织和血清中上调,是影响骨肉瘤预后的生物标志物;Hsa_circ_0005721在骨肉瘤患者血清中上调,可作为骨肉瘤无创性诊断分子标志物。

Expression level of circular RNA hsa_circ_0005721 in tissues and serum of patients with osteosarcoma and its clinical significance

Abstract

Objective: To explore the expression level of hsa_circ_0005721 in tumor tissues and serum samples of patients with osteosarcoma and its clinical significance. Methods: The relative expression levels of hsa_circ_0005721 in tumor tissues and adjacent normal tissues of 50 cases osteosarcoma patients were detected by qRT-PCR. The correlation between hsa_circ_0005721 expression levels and clinical pathological parameters was analyzed. Kaplan-Meier survival analysis were used to compared the Disease-free and Overall survival rate between hsa_circ_0005721 high and low expression patients. The relative expression levels of hsa_circ_0005721 in 30 cases of osteosarcoma, 30 cases of benign bone tumors and 30 cases of healthy people were detected by qRT-PCR. The diagnostic value of serum hsa_circ_0005721 in patients with osteosarcoma was investigated by ROC analysis. Results: The expression level of hsa_circ_0005721 in osteosarcoma was significantly higher than that in adjacent non-tumor tissues (P<0.01). The relative expression level of hsa_circ_0005721 was significantly correlated with Enneking stage and lung metastasis (P<0.01). The disease-free survival rate and overall survival rate of patients with high expression of hsa_circ_0005721 were significantly lower than those with low expression level of hsa_circ_0005721 (P<0.001). The expression of hsa_circ_0005721 was independent prognostic factor for disease-free survival (P=0.001) and overall survival (P=0.021) in osteosarcoma patients. The relative expression level of hsa_circ_0005721 in osteosarcoma patients was significantly higher than that in benign bone tumor patients and healthy controls (P<0.001). The relative expression level of hsa_circ_0005721 in osteosarcoma patients can differentiate osteosarcoma patients from healthy individuals with ROCAUC of 0.821 (95% CI 0.651–0.891), and differentiate osteosarcoma patients from benign bone tumors patients with a ROCAUC of 0.701 (95% CI 0.601–0.801). Conclusion: Hsa_circ_0005721 is up-regulated in osteosarcoma and is a good prognostic biomarker for osteosarcoma. Hsa_circ_0005721 is up-regulated in the serum of patients with osteosarcoma and can be used as a promising non-invasive biomarker for osteosarcoma.
