结肠癌组织中KiSS-1和FHL1的表达与临床意义

摘要
目的:研究结肠癌患者癌组织中人吻素1(kisspeptin 1,KiSS-1)、4个半LIM结构域蛋白1(four and a half LIM domains protein 1,FHL1)的表达与临床意义。方法:选择唐山市人民医院2010年10月至2011年5月收治的69例接受手术及术后化疗的结肠癌患者作为研究对象,分别采用免疫组织化学技术和RT-PCR技术检查KiSS-1与FHL1在结肠癌组织中的表达,随后结合患者临床信息及随访资料分析KiSS-1与FHL1表达在结肠癌中的临床意义。结果:结肠癌组织中KiSS-1,FHL1蛋白及mRNA均低于癌周组织,差异具有统计学意义(P<0.05);KiSS-1,FHL1表达与肿瘤分化程度、淋巴结转移有关(P<0.05);KiSS-1和FHL1阳性表达患者的5年总体生存率明显高于阴性表达患者,差异具有统计学意义(P<0.05);结肠癌组织中KiSS-1与FHL1表达呈正相关(r=0.698,P=0.012),均是结肠癌患者预后的独立影响因素(P<0.05)。结论:KiSS-1与FHL1在结肠癌组织中表达下降或缺失,联合检测KiSS-1与FHL1蛋白对判断结肠癌生物学行为、评估患者预后具有一定的临床价值。

Abstract
Objective: To study the expression and clinical significance of kisspeptin 1 (KiSS-1) and four and a half LIM domains protein 1 (FHL1) in colon cancer tissues. Methods: A total of 69 patients with colon cancer who had received operation and postoperative chemotherapy from October 2010 to May 2011 in Tangshan People’s Hospital were selected. The expressions of KiSS-1 and FHL1 in colon cancer tissues were detected by immunohistochemistry and RT-PCR. Combined with clinical information and follow-up data, the clinical significance of KiSS-1 and FHL1 expression in colon cancer was analyzed. Results: The protein and mRNA of KiSS-1 and FHL1 in colon cancer tissues were significantly lower than those in tissues around cancer (P<0.05). The expression of KiSS-1 and FHL1 were related to the degree of differentiation and lymph node metastasis (P<0.05). The 5-year overall survival rate of patients with positive KiSS-1 and FHL1 expression was significantly higher than that of patients with negative expression, and the difference was statistically significant (P<0.05). There was a significant positive correlation between expression of KiSS-1 and FHL1 (r=0.698, P=0.012), and the expression of KiSS-1 and FHL1 were independent prognostic factors of colon cancer patients (P<0.05). Conclusion: KiSS-1 and FHL1 are down-expressed or absent in colon cancer tissues. The combined detection of KiSS-1 and FHL1 protein has certain clinical value in judging the biological behavior of colon cancer and evaluating the prognosis of patients with colon cancer.
