摘要

目的:探讨miR-146b在甲状腺乳头状癌中的表达及其对甲状腺乳头状癌侵袭转移的影响。方法:体外培养人甲状腺乳头状癌TPC-1细胞并转染miR-146b,分为4组:miR-146b抑制组及抑制对照组、miR-146b转染组及转染对照组。分别通过qRT-PCR检测miR-146b的表达,CCK-8法检测细胞增殖,Transwell法检测细胞侵袭能力,划痕法检测细胞的迁移能力。结果:MiR-146b在甲状腺乳头状癌TPC-1细胞中高表达(P<0.01)。CCK-8法检测发现miR-146b抑制组TPC-1细胞活力与抑制对照组相比显著降低(P<0.01),而miR-146b转染组细胞活力与转染对照组相比显著提高(P<0.01)。Transwell法检测发现抑制miR-146b后细胞的穿膜数明显减少(P<0.01),而过表达miR-146b后细胞的穿膜数明显增加(P<0.01)。划痕法检测发现miR-146b抑制组的迁移距离与抑制对照组比显著扩大(P<0.01),而miR-146b转染组的迁移距离与对照组比显著缩小(P<0.01)。结论:甲状腺乳头状癌TPC-1细胞中存在miR-146b的高表达。miR-146b的高表达可促进TPC-1细胞的增殖、侵袭及迁移。

Expression of miR-146b in thyroid cancer and its role in invasion and migration of thyroid cancer cells

Abstract

Objective: To investigate the expression level of micro RNA-146b in thyroid cancer cell lines and its impact on thyroid cancer invasion and metastasis. Methods: MiR-146b was transfected into human thyroid cancer cells TPC-1 in vitro. All cells were divided into an inhibited miR-146b group, an inhibitor negative control group, a transfected miR-146b group, and a negative control group. MiR-146b expression was detected by qRT-PCR in thyroid cancer cell lines (TPC-1) and negative control cell lines. The characteristics of invasion and migration of the transfected MiR-146b cells were examined by transwell assay. The expression of miR-146b was detected by qRT-PCR, cell proliferation was determined by CCK-8 assay. Migration rate was measured by wound healing assay. Results: MiR-146b was highly expression in thyroid cancer cells TPC-1 (P<0.01). CCK-8 assay found that the cell viability in the miR-146b inhibitor group were significantly lower than that in the inhibitor NC group (P<0.01), while that in the miR-146b mimics group were significantly higher than that in the NC group (P<0.01). Transwell assay found that the number of cells through membranes was decreased evidently in the miR-146b inhibitor group (P<0.01), while that was increased evidently in the miR-146b mimics group (P<0.01). The wound healing assay found that the migration distance of miR-146b inhibitor group was farther than that in the inhibitor NC group (P<0.01), while that of the miR-146 mimics group was shorter than that in the NC group (P<0.01). Conclusion: MiR-146b has high expression in thyroid cancer cells TPC-1. The overexpression miR-146b can promote cell proliferation, invasion, and migration.
