摘要

目的:研究分拣蛋白相关受体-1(sortilin-related receptor 1,SORL1)基因和胰岛素样生长因子-1(insulin-like growth factor-1,IGF-1)在老年糖尿病合并阿尔茨海默病(Alzheimer’s disease,AD)患者中的表达及相关性分析。方法:选择30例老年糖尿病AD患者(A组)、30例老年AD不合并糖尿病患者(B组)、30例老年糖尿病不合并AD患者(C组)和30例老年健康人群(D组),采用PCR法检测血清SORL1 mRNA水平,ELISA法检测血清IGF-1和β淀粉样多肽(Aβ)水平,免疫组织化学染色检测微管结合蛋白(Tau)磷酸化水平;比较各组上述指标表达的差异性,分析SORL1 mRNA和IGF-1水平的相关性。结果:A组血清SORL1 mRNA水平最高,B组次之(P<0.05);C组和D组差异无统计学意义(P>0.05)。血清IGF-1和Aβ水平、Tau磷酸化阳性率A组>B组>C组>D组,差异有统计学意义(P<0.05)。Pearson检验显示:A组SORL1 mRNA与IGF-1水平呈负相关(r=−0.723,P=0.013),其他3组无相关性(P>0.05)。结论:SORL1基因和IGF-1在老年糖尿病合并AD患者中异常表达,可影响Aβ和Tau蛋白的形成;结果还需要进一步验证。

Expressions of sortilin-related receptor 1 gene and insulin-like growth factor 1 and its correlation in elderly diabetic and Alzheimer's disease patients

Abstract

Objective: To study the expressions of sortilin-related receptor 1 (SORL1) gene and insulin-like growth factor-1 (IGF-1) in elderly diabetic and Alzheimer’s disease (AD) patients and to analyze the correlation of the levels of SORL1 gene and IGF-1. Methods: A total of 30 elderly diabetic and AD patients were enrolled in group A, and 30 elderly AD without diabetic patients were enrolled in group B, and 30 elderly diabetic without AD patients were enrolled in group C, and the other 30 elderly healthy people were enrolled in group D. The serum levels of SORL1 mRNA were detected by PCR, and the serum levels of IGF-1 and Aβ were detected by ELISA. The expressions of microtubule-binding protein (Tau) phosphorylation were detected by immunohistochemical staining. All above indexes were compared among four groups. The correlations of the levels of SORL1 gene and IGF-1 in four groups were analyzed by Pearson test. Results: The levels of SORL1 mRNA were different among four groups, and the sequences were group A > group B > group C > group D (all P<0.05). The levels of IGF-1 and Aβ and the positive rate of Tau protein phosphorylation were different among four groups, and the group A was highest (all P<0.05), the group B was the second highest, but there was no difference between the group C and D (P>0.05). By Pearson test, there was negative correlation between SORL1 mRNA and IGF-1 levels in group A (r=−0.723, P=0.013), but there were no correlation between SORL1 mRNA and IGF-1 levels in other three groups (P>0.05). Conclusion: The abnormal expressions of SORL1 gene and IGF-1 in elderly diabetes and AD patients can influence Aβ and Tau proteins formation, which need further study to prove it.
