细胞周期调控系统Cyclins-CDKs-CKIs在创面愈合中作用的研究进展

摘要

细胞周期调控系统相关因子调控细胞周期进展,在创面愈合炎症期与修复期均发挥重要作用。细胞周期蛋白(Cyclin)通过Cyclin依赖性激酶(Cyclin-dependent kinases,CDKs)调控细胞周期,而CDKs的抑制剂(Cyclin-dependent kinase inhibitor,CKI)具有抗炎作用,在炎症期抑制cyclinD1基因的表达可能通过防止巨噬细胞过度增殖,从而消退炎症反应,利于慢性创面的愈合;在创面修复期下调细胞周期正向调控因子CyclinD,CyclinE1,上调细胞周期负向调控因子p21cip1,p27kip1等的表达可抑制成纤维细胞的异常增殖从而减少瘢痕增生。Cyclins-CDKs-CKIs的潜在治疗价值是消退炎症反应,促进创面愈合,并抑制瘢痕增生,深入了解其在创面愈合不同阶段中的作用,对探索促进创面愈合、抑制瘢痕增生的治疗靶点具有重要意义。

Abstract

Cell cycle regulation system-related factors regulate cell cycle progression, and have important expression significance in wound healing and repair phase. Cyclin regulates cell cycle through cyclin-dependent kinases (CDKs), and inhibitors of CDKs have anti-inflammatory effects. Inhibition of CyclinD1 gene expression during inflammatory phase can prevent hyper proliferation of macrophages, thereby may attenuating inflammatory response and facilitating healing of chronic wounds; Down-regulation of cell cycle positive regulators CyclinD, CyclinE1, upregulation of cell cycle negative regulatory factors p21cip1, p27kip1 can inhibit the abnormal proliferation of fibroblasts and reduce scar hyperplasia. The potential therapeutic value of Cyclins-CDKs-CKIs is to attenuate the inflammatory response to promote wound healing and reduce scar hyperplasia, and to understand its role in different stages of wound healing. It is of great significance for exploring therapeutic targets for promoting wound healing and inhibiting the development of hypertrophic scars.
