糖尿病前期和2型糖尿病患者血清中miR-29b的表达水平及临床意义

摘要

目的:研究血清中miR-29b的表达与糖尿病前期和2型糖尿病(type 2 diabetes mellitus,T2DM)的相关性以及作为辅助诊断的生物标志物潜能。方法:采用生化分析仪检测相关临床生化指标;qRT-PCR检测对照组,糖尿病前期组和T2DM组血清中miR-29b的表达水平;分析糖尿病前期组和T2DM组血清中miR-29b与纤维蛋白原(fibrinogen,FG),HbA1c和口服葡萄糖耐量试验(oral glucose tolerance test,OGTT)的相关性;logistic回归分析血清中miR-29b与糖尿病前期和T2DM的相关性;ROC曲线分析评估血清中miR-29b诊断糖尿病前期和T2DM的价值。结果:miR-29b在T2DM组和糖尿病前期组血清中的表达水平明显高于对照组(P<0.05),相比糖尿病前期组,miR-29b在T2DM组血清中表达水平明显升高(P<0.05);糖尿病前期组和T2DM组血清中miR-29b与FG,HbA1c和OGTT均呈正相关;血清中miR-29b是糖尿病前期和T2DM的独立危险因素;血清中miR-29b诊断糖尿病前期和T2DM的ROC曲线下面积(AUC)分别为(AUC=0.681;95%CI 0.587~0.776)和(AUC=0.757,95%CI 0.673~0.841)。结论:血清中miR-29b的表达水平是潜在的诊断糖尿病前期及T2DM的生物标志物。

Expression level and clinical significance of miR-29b in serum of pre-diabetes and T2DM patients

Abstract

Objective: To investigate the correlation between the expression of miR-29b in serum with the prediabetes and type 2 diabetes mellitus (T2DM), and its potential as a biomarker for auxiliary diagnosis. Methods: Relevant clinical biochemical indexes were detected by biochemical analyzer. The expression level of miR-29b in the serum of 3 groups was detected by qRT-PCR. The correlation of miR-29b and fibrinogen (FG) were analyzed in prediabetes and T2DM groups, as the same as HbA1c and oral glucose tolerance test (OGTT); Logistic regression was used to the correlation of serum miR-29b and pre-diabetes and T2DM; ROC curve analysis was used to assess value of serum miR-29b as the diagnosis of pre-diabetes and T2DM. Results: The expression of miR-29b in serum of T2DM group and pre-diabetic group was significantly higher than that of control group (P<0.05). Compared with pre-diabetic group, the expression of miR-29b was significantly increased in T2DM group (P<0.05). There was a positive correlation between miR-29b, FG, HbA1c and OGTT in the prediabetes and T2DM groups; serum miR-29b was an independent risk factor for prediabetes and T2DM. The area under the curve (AUC) of serum miR-29b as diagnose of prediabetes and T2DM were (AUC=0.681; 95% CI 0.587–0.776) and (AUC=0.757; 95% CI 0.673–0.841), respectively. Conclusion: The expression level of miR-29b in serum is a potential biomarker for the diagnosis of prediabetes and T2DM.
