下调miRNA-181a降低寒冷刺激诱导的妊娠高血压小鼠的血压

摘要

目的:检测miRNA-181a在寒冷刺激诱导的妊娠高血压小鼠胎盘组织中的表达,研究下调miRNA-181a表达后对妊娠高血压小鼠血压的影响,并探讨初步机制。方法:尾静脉注射200 nmol/kg miRNA-181a-antagomir,2 d后,通过反复寒冷应激刺激方法建立妊娠高血压小鼠模型。颈动脉插管法测量小鼠血压,qPCR检测胎盘组织中miRNA-181a的表达,使用NO试剂盒测量血清中NO含量,ELISA检测血清中血红素氧合酶-1(heme oxygenase 1,HO-1)含量,Western印迹法检测胎盘组织中Akt,pAkt,内皮型一氧化氮合酶(eNOS)和peNOS蛋白表达。结果:与对照组相比,寒冷刺激诱导的妊娠高血压小鼠胎盘组织中miRNA-181a水平显著增加。与模型组相比,沉默miRNA-181a能够降低孕鼠的血压,升高血清NO和HO-1水平,并上调胎盘组织中pAkt和peNOS蛋白表达。结论:miRNA-181a表达可能与寒冷刺激诱导的妊娠高血压的发生发展有潜在关系。

Down-regulation miRNA-181a reduces blood pressure in cold-induced pregnant hypertensive mice

Abstract

Objective: To examine the expression of miRNA-181a in placenta tissue of cold induced-pregnant hypertensive mice and to explore the effects of and the possible mechanisms of down-regulation miRNA-181a on blood pressure. Methods: Mice were injected with 200 nmol/kg miRNA-181a-antagomir via tail vein. Two days later, model of pregnant hypertension was established by cold-stress stimulation method. Blood pressure was measured with direct intubation of carotid. The expressions of miRNA-181a in placenta tissues were examined by qPCR; the levels of serum NO were determined by NO assay kit; the levels of serum heme oxygenase 1 (HO-1) were determined by ELISA assay; The protein expressions of Akt, pAkt, eNOS and peNOS were analyzed by Western blot. Results: Compared to control group, the expression of miRNA-181a in placenta tissue of cold induced-pregnant hypertensive mice was significantly increased. Moreover, compared to model group, inhibition of miRNA-181a suppressed the blood pressure, increased the levels of serum NO and HO-1, and further, increased the protein expression of pAkt and peNOS in placenta tissue, respectively. Conclusion: MiRNA-181a may be related to the development of cold induced-pregnant hypertension.
