信号素4D在结直肠癌组织中的表达及其与微血管密度的关系

摘要
目的:探讨结直肠癌组织中信号素4D(semaphorin4D,Sema4D)的表达及其与微血管密度(microvessel density,MVD)的相关性。方法:回顾性分析2014至2015年间在云南省肿瘤医院结直肠外科接受结直肠癌根治性手术的61例患者的临床病理资料;采用免疫组织化学SP法检测该组患者的结直肠癌组织标本及其56例正常肠黏膜组织中Sema4D蛋白的表达水平;采用微血管密度计数法计数用CD34标记的微血管。结果:Sema4D主要表达于结直肠癌细胞膜及细胞质,正常肠黏膜组织细胞质内呈阴性表达。癌组织Sema4D表达阳性率(61/61)明显高于正常肠黏膜组织(0/56),差异有统计学意义(χ2=113.027,P<0.001)。Sema4D与MVD两者作相关性分析,两者有显著的正相关(r=0.333,P=0.009)。MVD表达量在Sema4D表达高低组间存在显著差异(P=0.003)。结论:Sema4D在结直肠癌组织高水平表达,且与MVD存在明显的正相关性,提示其有望成为潜在的结直肠癌血管治疗靶点。

Expression of Semaphorin 4D in colorectal carcinoma and its relationship with microvessel density

Abstract
Objective: To investigate the expression of semaphorin 4D (Sema4D) in colorectal carcinoma and its correlation with microvessel density (MVD). Methods: We retrospectively analyzed the clinical and pathological data of 61 patients with colorectal cancer undergoing colorectal surgery from 2014 to 2015 in Yunnan Provincial Tumor Hospital. The expression levels of Sema4D protein in colorectal cancer tissues and 56 cases of normal intestinal mucosal tissue were detected by immunohistochemical SP method. MVD markers were used to count microvessels labeled with CD34. Results: Sema4D was mainly expressed in the cytoplasm and cytoplasm of colorectal carcinoma, and the expression of Sema4D was negative in the cytoplasm of the normal intestinal mucosal tissue. The positive rate of Sema4D expression in cancer tissues (61/61) was significantly higher than that in normal intestinal mucosal tissue (0/56), the difference was statistically significant (χ2=113.027, P<0.001). Sema4D and MVD were analyzed by Spearman correlation coefficient (r=0.333, P=0.009), which had significant positive correlation. T test analysis of MVD expression indicated significant differences between the high and low expression of Sema4D groups (P=0.003). Conclusion: Sema4D is highly expressed in colorectal cancer tissue, and there is a significant correlation with MVD, suggesting that it is potential to be a potential therapeutic target of colorectal cancer.
