摘要

目的:探讨外源性NO对CNE-2细胞增殖与凋亡的影响。方法:分别用不同浓度NO供体药物硝普钠(sodium nitroprusside,SNP)干预CNE-2细胞,观察细胞形态学变化、检测细胞的抑制率及细胞的凋亡和坏死率。结果:SNP以浓度依赖性方式抑制CNE-2细胞增殖,SNP 100,200,400,600,800,1 600,3 200 μmol/L各组细胞抑制率与药物浓度呈正相关;SNP以浓度依赖性方式促进CNE-2细胞凋亡,SNP(1 000 μmol/L)组细胞凋亡率较SNP(600 μmol/L)组显著增高(P<0.05)。结论:外源性NO能抑制CNE-2的增殖,促进CNE-2的凋亡,其抑制效应与NO浓度呈正相关。

Abstract

Objective: To investigate the effect of proliferation and apoptosis induced by exogenous NO on CNE-2 cells. Methods: CNE-2 cells were treated by different concentrations of sodium nitroprusside (SNP). The morphological changes of cells were observed, the viability and apoptosis of cells were examined. Results: SNP inhibited proliferation of CNE-2 cell in a concentration-dependent manner, CNE-2 cell were treated by SNP 100, 200, 400, 600, 800, 1 600, 3 200 μmol/L group respectively and the inhibition rate of the cells of each group was positively correlated with the concentration of SNP. SNP promote apoptosis of CNE-2 cell in a concentration-dependent manner; compared with SNP 600 μmol/L, flow cytometry analysis showed that the apoptosis rate of CNE-2 cells on the group treated by SNP 1 000 μmol/L increased significantly (P<0.05). Conclusion: Exogenous nitric oxide can inhibit CNE-2 proliferation and promote apoptosis. The inhibitory effect was positively correlated with the concentration of NO.
