摘要

目的:检测不同类型肺癌患者与健康人血清中DNA甲基化酶1(DNA methylase 1,DNMT1)和甲基胞嘧啶双加氧酶(ten-eleven translocation methylcytosine dioxygenase 1,TET1)的表达水平,探讨2种酶对肺癌早期诊断的临床意义。方法:选取170例初诊为肺癌的患者血清与90例体检中心健康成年人血清,采用酶联免疫吸附试验(enzyme-linked immunoadsordent assay,ELISA)检测血清中DNMT1与TET1的表达水平。使用SPSS 26.0与Medcalc软件进行统计分析,比较健康成年人与肺癌患者血清中DNMT1与TET1表达水平的差异;比较血清DNMT1和TET1表达水平在不同临床分期、不同类型肺癌、不同性别、不同年龄段、不同吸烟史、不同癌症家族史之间的差异。结果:肺癌患者血清中的DNMT1显著高于健康成年人,而TET1的表达水平低于健康成年人,差异均有统计学意义(均P<0.05)。两种酶在肺癌患者血清中的表达水平与肺癌患者的临床病理分期有关。此外,DNMT1与TET1表达水平与不同类型肺癌的分类有关。结论:DNMT1与TET1有作为肺癌早期诊断标志物的潜在价值。

Abstract

Objective: To detect the expression levels of DNA methylase 1 (DNMT1) and ten-eleven translocation methylcytosine dioxygenase 1 (TET1) in the serum of patients with different types of lung cancer and healthy people, and to evaluate the clinical significance of the 2 enzymes in the early diagnosis of lung cancer.
Methods: The serum of 170 patients with lung cancer and 90 healthy subjects were selected, and the serum expression levels of DNMT1 and TET1 were detected by enzyme-linked immunoadsordent assay (ELISA). SPSS 26.0 and Medcalc software were used for statistical analysis to compare the serum DNMT1 and TET1 levels in patients between lung cancer patients and healthy controls. The level of serum DNMT1 and TET1 was compared in different clinical stages, different types of lung cancer, different genders, different age groups, different smoking history, and different family history of cancer.
Results: The serum expression level of DNMT1 of lung cancer patients was significantly higher while TET1 was significantly lower than that of the healthy subjects (P<0.05); the expression levels of the 2 enzymes in the serum of lung cancer patients were related to the clinicopathological stage; in addition, DNMT1 and TET1 have high sensitivity in the lung cancer diagnosis and may be related to the classification of different types of lung cancer.
Conclusion: DNMT1 and TET1 have potential value as early diagnostic markers of lung cancer.
