泛素-蛋白酶体途径异常调控在吗啡耐受及依赖分子机制中的作用

摘要
吗啡是目前治疗中、重度疼痛最常用和有效的阿片类药物。但是,长期反复使用吗啡容易产生药物耐受和依赖,这在很大程度上限制了吗啡的临床应用。泛素-蛋白酶体途径是细胞内蛋白质降解的主要途径,在多种细胞生命过程中发挥重要作用。研究表明泛素-蛋白酶体途径异常调控可能是吗啡耐受及依赖发生的重要机制之一,本文就此方面的研究进展作一综述。深入阐明吗啡诱导泛素–蛋白酶体途径异常调控的具体机制及其与中枢神经适应性改变之间的关系,对于进一步揭示吗啡耐受及依赖分子机制具有十分重要的意义。

The role of ubiquitin-proteasome pathway in the molecular mechanism of morphine tolerance and dependence

Abstract
Morphine is a typical opioid drug mostly used for relieving moderate to serious pain in the clinical. However, repeated or prolonged administration of morphine would develop antinociptive tolerance and drug dependence, thus greatly limiting its clinical application. Ubiquitin-proteasome pathway (UPP) is a major proteolytic pathway in the cell that plays a key role in many cellular processes. Studies have indicated that dysregulation of UPP might be an important mechanism in the development of morphine tolerance and dependence, thus, we will summarize the research progress in this paper. Understanding the effect of morphine on the regulation of UPP and its relationship with morphine-induced neural adaptive changes in the central nervous system is of great significance for further revealing the molecular mechanism of morphine tolerance and dependence.
