高表达Bcl-2下调喉癌细胞黏附能力的分子机制探讨

摘要
目的:探讨高表达的Bcl-2对喉癌细胞黏附能力的影响。方法:建立并鉴定高表达Bcl-2的稳定细胞株,采用黏附实验检测细胞的黏附能力,流式细胞仪和Western blotting实验分析细胞表面integrin表达的变化。结果:成功构建高表达Bcl-2细胞株,下调了细胞的黏附能力,并降低了细胞表面integrin α2β1的表达,而未见对integrin αv、integrin β4、integrin α3和integrin α5的表达的影响。结论:高表达Bcl-2可能通过下调了细胞表面integrin α2β1的表达从而减低细胞的黏附能力。

Abstract
Objective: To investigate the effect of high expression of Bcl-2 on the cell adhesion ability. Methods: The stable cells of high expression of Bcl-2 were built and identified. The adhesion ability was assessed by adhesion assay. The expression of integrin was analyzed by FLOW and Western blotting. Results: Bcl-2 stable cells were successfully built. Bcl-2 overexpression decreased the cell adhesion and the expression of integrin α2β1, however, it did not affect the expression of integrin αv, integrin β4, integrin α3 and integrin α5. Conclusion: High expression of Bcl-2 may reduce the cell adhesion by down-regulating the expression of integrin α2β1.
