摘要

目的:探讨长链非编码RNA(long non-coding RNA,lncRNA)FGD5-AS1对人结直肠癌细胞株侵袭、迁移和凋亡能力的影响。方法:分别通过lncRNA FGD5-AS1干扰、过表达和阴性对照质粒转入结直肠癌细胞株(LoVo、HT29)。采用qRT-PCR检测转染效果;采用细胞流式法检测分析干扰、过表达lncRNA FGD5-AS1后对结直肠癌细胞LoVo和HT29凋亡能力的影响;采用细胞划痕试验分析干扰、过表达lncRNA FGD5-AS1后对结直肠癌细胞LoVo和HT29迁移能力的影响;采用Transwell实验分析干扰、过表达lncRNA FGD5-AS1后对结直肠癌细胞LoVo和HT29侵袭能力的影响。结果:成功构建干扰、过表达lncRNA FGD5-AS1的结直肠癌稳转细胞株,转染lncRNA FGD5-AS1干扰质粒的结直肠癌细胞LoVo和HT29侵袭、迁移能力显著下降,凋亡能力显著上升;转染lncRNA FGD5-AS1过表达质粒的结直肠癌细胞LoVo和HT29侵袭、迁移能力显著上升,凋亡能力显著下降。结论:LncRNA FGD5-AS1可显著提升结直肠癌细胞的侵袭及迁移能力,同时可显著抑制结直肠癌细胞的凋亡。

Abstract

Objective: To investigate the effects of long non-coding RNA (lncRNA) FGD5-AS1 on invasion, migration and apoptosis of human colorectal cancer cell lines (LoVo, HT29). Methods: LncRNA FGD5-AS1 was transfected into colorectal cancer cell lines (LoVo, HT29) by interference, overexpression and negative control plasmid. The transfection effect was detected by qRT-PCR assay. The effects of interference and overexpression of lncRNA FGD5-AS1 on the apoptotic ability of LoVo and HT29 in colorectal cancer cells were analyzed by flow cytometry. Cell scratching assay was used to analyze the effects of interference and overexpression of lncRNA FGD5-AS1 on the migration ability of LoVo and HT29 in colorectal cancer cells. Transwell experiment was conducted to analyze the effects of interference and overexpression of lncRNA FGD5-AS1 on the invasion ability of LoVo and HT29 in colorectal cancer cells. Results: Stable colorectal cancer cell lines with interference and overexpression of lncRNA FGD5-AS1 were successfully constructed. The invasion and migration ability of LoVo and HT29 cells transfected with lncRNA FGD5-AS1 interference plasmid were significantly decreased, and the apoptosis ability was significantly increased. The invasion and migration ability of LoVo and HT29 cells transfected with LncRNA FGD5-AS1 overexpressing plasmid were significantly increased, while the apoptosis ability was significantly decreased. Conclusion: lncRNA FGD5-AS1 can significantly enhance the invasion and migration ability of colorectal cancer cells, and significantly inhibit the apoptosis of colorectal cancer cells.
