枸杞多糖对顺铂化疗诱导的大鼠卵巢早衰模型的卵巢保护作用

摘要
目的:探讨枸杞多糖(lycium barbarum polysaccharide,LBP)对顺铂诱导的大鼠卵巢早衰模型的作用。方法:将80只雌性SD大鼠随机分为对照组、单纯顺铂给药组(DDP组)、顺铂+低剂量LBP处理组(DDP+LBP-L组)、顺铂+高剂量LBP处理组(DDP+LBP-H组)4组。对照组大鼠正常饲养;DDP组大鼠每天腹腔注射2 mg/kg DDP,持续14 d;DDP+LBP-L组和DDP+LBP-H组大鼠每天分别灌胃5 mg/kg,10 mg/kg LBP溶液后,再腹腔注射2 mg/kg DDP,持续14 d。用药14 d后,采集股静脉血、卵巢组织和子宫组织。取卵巢组织进行HE染色和DDX4免疫组织化学染色;对卵巢各发育阶段的卵泡计数和形态进行分析;检测血清中卵泡刺激素(follicle-stimulating hormone,FSH)和雌二醇(estradiol,E2)的含量;取子宫组织进行HE染色。结果:与对照组相比,DDP组大鼠的卵巢受损,原始卵泡、初级卵泡、窦前卵泡、窦卵泡和总卵泡数减少,闭锁卵泡增加。而DDP+LBP组与对照组卵巢相似,其中高剂量LBP组相似程度更高;与对照组相比,DDP能升高血清中FSH含量而降低E2含量,LBP可改善此情况;DDP处理可损伤大鼠子宫内膜,使腺体数目减少,而LBP处理后子宫内膜损伤情况减轻,且高剂量LBP改善程度更明显。结论:顺铂可对大鼠卵巢造成结构和功能的损害,枸杞多糖可在一定程度上减轻这种损伤,对卵巢起保护作用。

Protective effect of lycium barbarum polysaccharide on premature ovarian failure induced by cisplatin chemotherapy in rats

Abstract
Objective: To investigate the effects of lycium barbarum polysaccharide (LBP) on cisplatin-induced premature ovarian failure in rats. Methods: A total of 80 female SD rats were randomly divided into four groups: a control group, a DDP alone group (DDP group), a DDP + low-dose LBP group (DDP + LBP-L group), and a DDP + high-dose LBP group (DDP + LBP-H group). The rats in the control group were raised normally. The rats in DDP group were intraperitoneally injected with 2 mg/kg DDP every day for 14 days. The rats in the DDP+LBP-L group and the DDP+LBP-H group were given 5 and 10 mg/kg LBP solution every day, respectively, followed by intraperitoneal injection of 2 mg/kg DDP for 14 days. After administration for 14 days, femoral venous blood, ovarian tissues and uterine tissues were collected. Ovarian tissues were taken for HE staining and DDX4 immunohistochemical staining. Follicle counts and morphology at different stages of ovarian development were analyzed. The contents of follicle-stimulating hormone (FSH) and estradiol (E2) in serum were detected. Uterine tissue was taken for HE staining. Results: Compared with the control group, in the DDP group, the ovary was damaged, the number of primordial follicles, primary follicles, presinus follicles, sinus follicles and total follicles was decreased, and the number of atresia follicles was increased. However, the ovary of the DDP + LBP group was similar to that of the control group, and the high-dose LBP group had higher degree of similarity. Compared with the control group, DDP increased the content of FSH and decrease the content of E2 in serum, and LBP could improve this situation. DDP treatment could damage the endometrium and reduce the number of glands, while the endometrial damage was reduced after LBP treatment, and the improvement degree of high dose LBP was more obvious. Conclusion: Cisplatin can damage the structure and function of the ovary of rats, while LBP can weaken the adverse effect and protect the ovary in some cases.
