大黄素对盲肠结扎穿孔脓毒症大鼠模型各器官氧化应激与炎症反应的保护作用

摘要

目的:观察从中药中提取的大黄素对盲肠结扎穿孔脓毒症大鼠模型各器官氧化应激与炎症反应损伤的保护作用。方法:将48只雄性Wistar大鼠随机分为假手术组、假手术+大黄素组、盲肠结扎穿孔脓毒症(cecal ligation puncture,CLP)模型组及大黄素治疗组4组,每组12只。本实验采用盲肠结扎穿刺(cecum ligation and puncture,CLP)方法构建成年雄性Wistar大鼠盲肠结扎穿刺脓毒症模型。在模型构建前30 min,分别向大鼠腹腔内注射生理盐水或大黄素(25 mg/kg),在CLP术后48 h处死大鼠。测定各组大鼠的心、肝、肺和肾的髓过氧化物酶(myeloperox idase,MPO)、脂质过氧化物(malondialdehyde,MDA)、超氧化物歧化酶(superoxide dismutase,SOD)和过氧化氢酶(catalase, CAT)的活性。通过HE染色观察各组大鼠心、肝、肺、肾组织中的病理变化。结果:与Sham组比较,CLP组心、肝和肺组织中MPO活性和MDA含量显著增加,SOD和CAT活性显著降低;CLP组肾组织中MPO活性显著增加,SOD活性显著降低,而MDA含量和CAT活性没有明显变化。与CLP组比较,大黄素治疗组的心、肝和肺组织中MPO活性和MDA含量显著降低,SOD和CAT活性显著增加;大黄素治疗组的肾组织中MPO含量、MDA、SOD和CAT活性没有明显变化。HE染色显示:CLP组大鼠的心、肝、肺肾组织炎性细胞浸润增多;而大黄素治疗组大鼠的心、肝、肺肾组织炎性细胞浸润减少。结论:大黄素对脓毒症大鼠心、肝和肺有保护作用,能在一定范围内预防脓毒症大鼠氧化应激与炎症反应。

Protective effect of emodin on oxidative stress and inflammation of various organs in rat model of cecum ligation perforation sepsis

Abstract

Objective: To evaluate the effect of emodin in different organs of rats submitted to an animal model of cecum ligation perforation sepsis. Methods: A total of 48 adult male Wistar rats were successfully recruited and divided into four groups, Sham group, Sham + emodin group, CLP group, and CLP + emodin group, 12 rats for each. This research contributes sepsis by cecal ligation puncture (CLP) procedure. The rats were intraperitoneally injected with saline or emodin (25 mg/kg) 30 minutes before the model construction, and sacrificed 48 hours after the CLP operation. The myeloperoxidase (MPO), malondialdehyde (MDA), superoxide dismutase (SOD), and catalase (CAT) activities were evaluated in the heart, liver, lung, and kidney. The pathological changes of heart, liver, lung and kidney tissues in each group were observed by HE staining. Results: Compared with Sham group, MPO activity and MDA content in heart, liver and lung tissues of CLP group were significantly increased, while SOD and CAT activities were significantly decreased; in kidney tissues of CLP group, MPO activity increased significantly, SOD activity decreased significantly, while MDA content and CAT activity were not significant changes. Compared with CLP group, MPO activity and MDA content in heart, liver and lung tissues of emodin treatment group were significantly decreased, while SOD and CAT activity were significantly increased. There were no significant changes of MPO content, and MDA, SOD and CAT activities in kidney tissues of the emodin treatment group. HE staining showed increased inflammatory cell infiltration in the heart, liver, lung and kidney tissues of rats in the CLP group; however, inflammatory cell infiltration in heart, liver, lung and kidney tissues of emodin treatment group was decreased. Conclusion: Emodin has a protective effect on heart, liver and lung of sepsis rats, and can prevent oxidative stress and inflammation in sepsis rats within a certain range.
