摘要

目的:探讨sur vivin基因在儿童胚胎性横纹肌肉瘤(embryonal rhabdomyosarcoma,ERMS)中的生物学作用。方法:采用sur vivin免疫组织化学检测ERMS肿瘤组织和正常骨骼肌组织的表达情况,小干扰RNA(siRNA)介导的sur vivin抑制对ERMS RD细胞凋亡的影响,原位末端标记法(TUNEL)检测RD细胞凋亡率,Western印迹法检测RD细胞caspase-3和caspase-9蛋白表达水平。结果:Sur vivin在ERMS中高表达而在正常骨骼肌组织中不表达,特异性siRNA干扰sur vivin表达导致RD细胞凋亡显著增加,sur v iv in表达的下调导致更高的凋亡率,sur v iv in表达的下调增加了caspase-3的活性,对caspase-9的活性无影响。结论:Survivin过表达可能通过抑制细胞凋亡而与ERMS发病机制有关。

Abstract

Objective: To investigate the biological roles of survivin in pediatric embryonal rhabdomyosarcoma (ERMS). Methods: Immunohistochemical stain with survivin-specific antibody were performed on primary ERMS tumor tissue and on normal skeletal muscle tissue. Subsequently, we explored the effect of small interfering RNA (siRNA)-mediated inhibition of survivin on the apoptosis potential in representative RD cell line. TUNEL method was used to detect the apoptosis of RD cell. The expression levels of caspase-3 and caspase-9 protein were measured by Western blot. Results: Survivin was highly expressed in ERMS primary tumor but not in normal skeletal muscle tissue. Disruption of survivin expression by survivin-specific siRNA resulted in a significant increase of apoptosis in human RD cell. Higher apoptotic rate was caused by the down-regulation of survivin expression. In addition, knockdown of survivin expression promoted the activation of caspase-3 but not caspase-9. Conclusion: Over-expression of survivin might be relevant to ERMS pathogenesis through inhibiting cell apoptosis in a caspase-dependent manner.
