基于数据挖掘分析非小细胞肺癌中RPA3的表达及其临床意义

摘要
目的:探索复制蛋白A(replication protein A,RPA)3在非小细胞肺癌(non-small cell lung cancer,NSCLC)中的表达及意义。方法:利用BioGPS数据库检索RPA3在正常组织中的表达;使用Oncomine数据库分析关于RPA3基因的信息,并对RPA3基因在肺癌中的表达进行荟萃分析;利用Kaplan-Meier Plotter数据库分析RPA3表达对NSCLC患者生存时间的影响;String-DB数据库分析与RPA3相互作用的蛋白。结果:RPA3在人体正常组织中均有表达。与正常肺组织相比,NSCLC组织中RPA3基因在mRNA水平呈高表达(P<1E-5)。RPA3基因的高表达与NSCLC患者的总体生存期(overall survival,OS)呈负相关(HR=1.48,P=2.2E-09)。亚组分析显示:RPA3高表达水平对肺腺癌患者OS有显著影响(HR=2.09,P=3.4E-09),而在鳞癌患者中,其表达水平对OS无显著影响(HR=0.97,P=0.77)。MCM2,ORC2,ORC3,PCNA,POLD1,PRIM1,RFC4,RPA1,RPA4等基因与RPA3有明显的相互作用。结论:通过肿瘤基因数据库信息挖掘表明,RPA3在NSCLC组织中呈高表达,且与患者预后相关,为深入研究RPA3在NSCLC发生发展中的作用提供重要理论依据。

Abstract
Objective: To explore the expression and significance of replication protein A (RPA) 3 in non-small cell lung cancer (NSCLC). Methods: Utilizing BioGPS database analyzed RPA3 expression in normal tissues. Using Oncomine database mined the expression of RPA3 in NSCLC patients. Exploiting Kaplan-Meier Plotter database studied the prognostic roles of RPA3 in NSCLC. The RPA3-interacting proteins were analyzed by String-DB database. Results: The BioGPS database shows RPA3 expression in all the human normal tissues. Compared with normal lung tissue, RPA3 gene highly expressed in NSCLC tissue at mRNA level (P<1E-5). The expression level of RPA3 gene was negatively correlated with overall survival (OS) of NSCLC patients (HR=1.48, P=2.2E-09). Subgroup analysis showed that RPA3 expression level had a significant effect on OS of patients with lung adenocarcinoma (HR=2.09, P=3.4E-09), while in squamous cell carcinoma its expression level had no significant effect on OS (HR=0.97, P=0.77). The MCM2, ORC2, ORC3, PCNA, POLD1, PRIM1, RFC4, RPA1, RPA4 and other genes have obvious interaction with RPA3. Conclusion: According to the information mining of tumor gene database, RPA3 mRNA was found highly expressed in NSCLC tissue and the expression of RPA3 has a significant impact on survival of NSCLC patients, which provides an important theoretical basis for further studying the role of RPA3 in the occurrence and development of NSCLC.
