摘要

目的:探究成髓细胞瘤癌基因(myeloblastoma oncogene,MYB)在舌鳞癌发生、发展中的作用以及相关作用机制。方法:通过荧光定量PCR(real-time PCR,qPCR)检测MYB在舌鳞癌细胞系(UM1,CAL-27,SCC-9)以及正常人口腔黏膜成纤维细胞(oral mucosal fibroblasts,OMFbs)中的表达情况。转染siRNA-MYB至SCC-9细胞中,采用四甲基偶氮唑(MTT)和Transwell法检测其对细胞活性、侵袭、迁移的影响。Western印迹观察核因子-κB(nuclear factor-κB,NF-κB)信号通路的变化。结果:MYB在舌鳞癌细胞系(UM1,CAL-27,SCC-9)细胞中的表达量明显高于正常人OMFbs(P<0.05)。沉默MYB可显著抑制SCC-9细胞的活性、侵袭、迁移(P<0.05)。Western印迹显示沉默MYB表达可显著抑制NF-κB磷酸化,降低基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)的表达量。结论:沉默MYB表达可通过NF-κB信号抑制舌鳞癌细胞细胞活性、侵袭、迁移能力。

Silencing MYB gene regulating the inhibitory effect of NF-κB signaling pathway on tongue squamous cell growth

Abstract

Objective: To investigate the role and molecular mechanism of myeloblastoma oncogene (MYB) in the development of tongue squamous cell carcinoma. Methods: The expression of MYB in tongue squamous cell carcinoma cell lines (UM1, CAL-27, SCC-9) and normal human oral mucosal fibroblasts (OMFbs) were detected by real-time PCR (qPCR). The cell activity and invasion and migration were analyzed by MTT and Transwell after being transfected with siRNA-MYB. Western blot was used to observe the changes of nuclear factor-κB (NF-κB) signaling pathway. Results: The expression of MYB in tongue squamous cell carcinoma cell lines (UM1, CAL-27, SCC-9) were significantly higher compared with normal human oral mucosa fibroblasts (P<0.05). Silencing MYB significantly inhibited the activity, invasion and migration of SCC-9 cells (P<0.05). Western blot analysis showed that silencing MYB expression significantly inhibited NF-κB phosphorylation and decreased matrix metalloproteinase-9 (MMP-9) expression. Conclusion: Silencing MYB expression can inhibit the activity, invasion and migration of tongue squamous cell carcinoma cells by inhibiting NF-κB signaling.
