Sfrp5,Klotho血清表达量在糖尿病患者氧化应激、脂肪细胞因子分泌中的相关性

摘要
目的:探究糖尿病患者的分泌型卷曲蛋白相关蛋白-5(secreted frizzled-related protein-5,Sfrp5),Klotho血清表达量以及与氧化应激、脂肪细胞因子分泌的相关性。方法:选取2016年1月至2017年11月首次确诊的60例2型糖尿病患者(T2DM组)和60例糖耐量减低患者(IGT组),同时招募60例同年龄段的健康体检者作为对照组。比较3组Sfrp5,Klotho血清表达量,氧化应激指标(超氧化物歧化酶、丙二醛)以及脂肪细胞因子(内脂素、脂联素和瘦素)水平差异,应用Person相关分析探究糖尿病患者Sfrp5,Klotho血清表达量与氧化应激、脂肪细胞因子的相关性。结果:IGT组和T2DM组的Sfrp5和Klotho的表达量明显低于对照组(P<0.05);T2DM组患者的Sfrp5和Klotho的表达量明显低于IGT组(P<0.05)。与对照组和IGT组相比,T2DM组的SOD、内脂素素和脂联素水平更低(P<0.05),而MDA和瘦素水平较高(P<0.05)。相关性分析显示:Sfrp5和Klotho的表达量与SOD(r=0.470,0.482)、内脂素(r=0.495,0.396)和脂联素(r=0.556,0.479)水平呈正相关,而与MDA(r=−0.567,−0.496)和瘦素(r=−0.552,−0.498)水平呈负相关。结论:糖尿病患者Sfrp5,Klotho血清表达量明显降低;Sfrp5,Klotho可能在糖尿病患者氧化应激、脂肪组织炎症反应中发挥重要作用。

Abstract
Objective: To investigate the relationship of Sfrp5 and Klotho serum levels on oxidative stress and secretion of adipocytokines in diabetic patients. Methods: Sixty patients with type 2 diabetes (T2DM group) and 60 patients with impaired glucose tolerance (IGT group) were selected for the first time from January 2016 to November 2017. Sixty healthy subjects of the same age were recruited as the control group. The serum levels of Sfrp5 and Klotho, oxidative stress indicators (superoxide dismutase, malondialdehyde) and adipocytokines (visfatin, adiponectin and leptin) were compared among the three groups. The Person correlation analysis was used to investigate the relationship between serum Sfrp5 and Klotho expression and oxidative stress and adipocytokines in diabetic patients. Results: The expressions of Sfrp5 and Klotho in the IGT group and the T2DM group were significantly lower than those in the control group (P<0.05). The expression of Sfrp5 and Klotho in the T2DM group were significantly lower than those in the IGT group (P<0.05). Patients with T2DM had lower levels of SOD, lipotropin and adiponectin (P<0.05) compared with the control group and the IGT group, while the MDA and Leptin levels were higher (P<0.05). Correlation analysis showed that the expression of Sfrp5 and Klotho positively correlated with the levels of SOD (r=0.470, 0.482), visfatin (r=0.495, 0.396) and adiponectin (r=0.556, 0.479), while the MDA (r=−0.567, −0.496) and leptin (r=−0.552, −0.498) levels were negatively correlated. Conclusion: The serum levels of Sfrp5 and Klotho are significantly decreased in diabetic patients, and Sfrp5 and Klotho may play important roles in oxidative stress and adipose tissue inflammation in diabetic patients.
