摘要
目的:探讨CD40 rs1535045多态性与海南汉族人群肺癌的相关性。方法:采用病例对照研究的方法,收集258例肺癌患者和270例健康对照者的外周静脉血,提取血液基因组DNA,应用等位基因特异性聚合酶链反应(allele-specific PCR,AS-PCR)法和焦磷酸测序法进行基因分型。结果:在非吸烟女性组中,C等位基因的分布频率高于对照组(P=0.001),相对于T等位基因,携带C等位基因的非吸烟女性的肺腺癌发病风险增加(OR=2.58,95% CI 1.50~4.45);吸烟男性组中,C等位基因的分布频率高于T等位基因(P=0.043),携带C等位基因吸烟男性的肺鳞癌发病风险增高(OR=1.93,95% CI 1.02~3.65)。结论:CD40 rs1535045多态性与肺癌的发生有关,对吸烟男性的肺鳞癌和非吸烟女性的肺腺癌的发生可能具有预测作用。

Abstract
Objective: To evaluate the association between the polymorphisms in CD40 rs1535045 and lung cancer.
Methods: We performed a case-control study with 258 patients and 270 controls. Genomic DNA was extracted using a blood genome extraction kit. Genotyping was conducted using allele-specific polymerase chain reaction (allele-specific PCR, AS-PCR) and pyrosequencing. Results: The frequency of the C allele of rs1535045 was significantly higher in non-smoking female patients with lung adenocarcinoma (P=0.001), compared with T allele, individuals with C allele had a higher risk for lung adenocarcinoma in non-smoking female (OR=2.58, 95% CI 1.50–4.45). The frequency of the C allele was also significantly higher in smoking man patients with lung squamous cell carcinomas (P=0.043), individuals with C allele had a higher risk with lung squamous cell carcinomas (OR=1.93, 95% CI 1.02–3.65). Conclusion: We find that the polymorphisms of CD40 rs1535045 were associated with the risk for lung cancer, and the polymorphisms of CD40 rs1535045 may be a predictor for lung adenocarcinoma in non-smoking female and squamous cell carcinomas in smoking man.
