乳腺癌中EZH2,MMP-2和MMP-9蛋白表达及其临床意义

摘要
目的:探讨组蛋白甲基转移酶(enhancer of zeste homolog 2,EZH2),MMP-2和MMP-9在乳腺癌组织中的表达,并分析三者的表达与乳腺癌临床病理特征的关系。方法:采用免疫组织化学SP法检测147例乳腺癌、58例正常乳腺组织和58例乳腺良性肿瘤组织中EZH2,MMP-2和MMP-9蛋白表达,分析三者间的关系。并将三者的表达与乳腺癌的临床病理特征进行相关性分析。结果:EZH2,MMP-2和MMP-9蛋白在乳腺癌组织中的表达显著高于正常乳腺组织及乳腺良性肿瘤组织(P均<0.001)。EZH2,MMP-2和MMP-9蛋白表达与乳腺癌患者年龄、绝经状态、肿瘤直径及组织学分类均无关,而与患者淋巴结转移有关(P<0.001)。结论:EZH2,MMP-2和MMP-9可能与乳腺癌的发生发展密切相关,三者联合检测可为乳腺癌的诊疗分析研究提供新的参考指标。

Abstract
Objective: To explore the expression of enhancer of zeste homolog 2 (EZH2), MMP-2 and MMP-9 in breast cancer and to analyze their relationship with the clinical pathologic characteristics. Methods: The expression of EZH2, MMP-2 and MMP-9 protein were detected by SP immunohistochemical method in 147 breast cancer tissues, 58 normal breast tissues and 58 benign breast tumor tissues, and the relationship among the three groups were analysed. The correlation of the EZH2, MMP-2 and MMP-9 protein expression with the clinicopathological features were also analyzed. Results: Positive intensity of EZH2, MMP-2 and MMP-9 in breast cancer were significantly higher than that in normal breast tissues and benign breast tumor (P<0.001). In breast cancer the expression of EZH2, MMP-2 and MMP-9 were not correlated with patient age, menopausal status, the tumor size and histological types, but were correlated with the lymph node metastasis (P<0.001). Conclusion: EZH2, MMP-2 and MMP-9 may be closely related to the pathogenesis and progression of breast cancer. The joint detection of the 3 indictors of EZH2, MMP-2 and MMP-9 in human breast cancer tissue may contribute to cancer diagnosis and treatment, and provide a reference to the study of breast cancer.
