己糖激酶Ⅱ与Warburg效应

摘要

肿瘤细胞在氧气充足的情况下通过葡萄糖的无氧氧化代谢方式为细胞提供能量的现象称为Warburg效应。己糖激酶是细胞糖酵解中的第一个限速酶,其中己糖激酶-Ⅱ与肿瘤细胞的糖酵解代谢、细胞增殖和凋亡等功能有着更为密切的联系。在肿瘤细胞中,基因水平的改变,信号通路的影响,促使己糖激酶高表达和活性增加,保证了肿瘤细胞糖酵解的快速进行。己糖激酶可通过与线粒体外膜结合促进糖酵解、抑制细胞凋亡和调节细胞自噬影响肿瘤细胞的功能。己糖激酶有望成为肿瘤治疗中的重要靶点。


Hexokinase-Ⅱ and Warburg effect

Abstract

The tumor cells provide energy through anaerobic oxidation of glucose in the case of sufficient oxygen. This phenomenon is known as the Warburg effect. Hexokinase is the first rate-limiting enzyme in the process of cell glycolysis, and Hexokinase-Ⅱ is linked more closely with tumor glycolysis, cell proliferation, apoptosis, etc. In tumor cells, the changes of gene and the influence of the signal path make the content of hexokinase significantly increased and activity increased, ensure the proceed of glycolysis in tumor cells. Hexokinase combines with the mitochondrial outer membrane to promote glycolysis, inhibit apoptosis and regulate autophagy to affect the function of tumor cells. Hexokinase is an important target in tumor treatment.
