摘要

氧化应激在帕金森病多巴胺能神经元变性中起着重要作用,活性氧(reactive oxygen species,ROS)产生的来源和机制包括多巴胺本身的代谢,线粒体功能障碍等。DJ-1在帕金森病和心力衰竭中起抗氧化作用,参与氧化应激保护细胞。DJ-1是癌症相关蛋白,它参与细胞内不同的信号通路。DJ-1可以调控氧化应激反应,对肿瘤细胞的侵袭转移行为产生了明确的影响,因此可以推测,DJ-1作为一种抗氧化剂可以影响肿瘤的治疗和复发。现在已证实DJ-1与癌症之间有着密切关系,但是它是如何作用的而改变细胞存活的信号通路的详细机制仍然未知。本文综述DJ-1在帕金森病,心衰,肿瘤中抗氧化机制,以及该靶点在肿瘤治疗中的研究进展。

Abstract

Oxidative stress have an important role in the degeneration of dopaminergic neurons in Parkinson’s disease, and the origin and mechanism of ROS (reactive oxygen species, ROS), including the metabolism of dopamine and mitochondrial dysfunction. DJ-1 plays an antioxidant role in Parkinson’s disease and heart failure, which is involved in oxidative stress and protecting cells. DJ-1 is a tumor-associated protein, which is involved in different cell signaling pathways. DJ-1 can regulate the oxidative stress, which has a definite effect on the invasion and metastasis of tumor cells, thus it can be speculated that DJ-1 acts as an antioxidant which can affect the treatment and recurrence of tumor. It has now been confirmed that there is a close relationship between DJ-1 and cancer, but it is still unclear how it works and the mechanism of signaling pathways that alter cell survival. In this paper, we reviewed the progress of DJ-1 in the treatment of Parkinson’s disease, heart failure, tumor, anti-oxidative mechanism, and the target in tumor therapy.
