JAK1/STAT1在EGFR 19-Del之NSCLC中的表达及其相关性研究

摘要

目的:检测信号分子p-JAK1、p-STAT1在EGFR外显子19缺失突变(以下简称EGFR 19-Del)的非小细胞肺癌(non-small-cell lung cancer,NSCLC)组织样本中的表达,探讨EGFR突变后下游信号通路JAK/STAT表达之间的相关性,为进一步研究EGFR 19-Del的NSCLC发生发展机制提供依据。方法:收集经实时荧光定量PCR——ARMS法筛选的125例EGFR 19-Del的NSCLC组织,应用免疫组织化学法检测该组织样本中p-JAK1和p-STAT1的表达,统计分析二者在不同临床病理特点中的表达差异以及表达相关性。结果:p-JAK1、p-STAT1阳性表达主要定位于细胞质和细胞核中。在125例EGFR 19-Del的NSCLC组织样本中,p-JAK1、p-STAT1阳性率分别为83.2%、100%。p-JAK1、p-STAT1在鳞癌、腺癌两组间的表达无统计学差异(P=0.440;P=0.822);p-JAK1、p-STAT1在总的NSCLC、腺癌的不同病理分级、TNM分期组间差异显著(P值均<0.01),而二者在鳞癌的不同病理分级、TNM分期组间无统计学差异(P值均>0.05)。p-JAK1与p-STAT1在总的NSCLC、腺癌、鳞癌中的表达率和表达水平存在相关性(P值均<0.05,rs值均>0.428);二者在病理分级的高、中分化组中有很好的相关性(P值均<0.01,rs值均>0.525),而在低分化组中的相关性不大(P>0.05);二者在TNM分期组中的早、晚期均有很好的相关性(rs=0.567,P=0.000;rs=0.477,P=0.000)。结论:p-JAK1、p-STAT1在EGFR 19-Del NSCLC组织样本中有很高的表达率,且p-JAK1、p-STAT1在病理分类、病理分级及TNM分期组间的表达有很好的相关性,说明JAK1/STAT1信号通路很可能与EGFR 19-Del的NSCLC发生发展有关,为进一步研究EGFR 19-Del之NSCLC发生发展机制提供重要依据。

The study on expression of JAK1/STAT1 and its correlation in NSCLC with EGFR exon 19 deletion

Abstract

Objective: The expression of p-JAK1, p-STAT1 was investigate and the correlation of p-JAK1 expression with p-STAT1 was analyzed in NSCLC with EGFR 19-Del. Methods: A total of 125 NSCLC samples with EGFR 19-Del were selected by ARMS, p-JAK1 and p-STAT1 were detected in those samples using immunochemistry and the correlation of their expression were statistically analyzed. Results: The expression of p-JAK1, p-STAT1 was mainly located in cytoplasm and nucleus. In those cases, the expression rates of p-JAK1, p-STAT1 protein were 83.2% and 100%. There were no significant differences of p-JAK1 and p-STAT1 expression level between the squamous carcinoma group and the adenocarcinoma group (P=0.440; P=0.822). There were significant differences of p-JAK1, p-STAT1 among the different pathological grades and TNM stages in the NSCLC and the adenocarcinoma (P<0.01), while there was no statistical difference in squamous carcinoma (P>0.05). The correlation of p-JAK1 with p-STAT1 for the expression rate and the level were obvious (P<0.05, rs>0.428) in the NSCLC, adenocarcinoma and squamous cell carcinomas; The correlations of p-JAK1 with p-STAT1 expression were also obvious in well and middle differentiation groups (P<0.01, rs>0.525), but little correlation in low differentiation group of NSCLC (P>0.05); Also there were good correlations in the TNM staging Ⅰ~Ⅱ, Ⅲ~Ⅳ groups in NSCLC. Conclusion: The p-JAK1, p-STAT1 protein were highly expressed in NSCLC with EGFR 19-Del, and there were good correlations of p-JAK1 expression with p-STAT1 in the pathological classification, pathologic stages and TNM stages, which suggest that JAK1/STAT1 signal pathway may be associated with the development of NSCLC, all that would provide important basis for the further study on the mechanism of NSCLC with EGFR 19 deletion mutation.
