采用抗体芯片筛选鼻咽癌放射治疗抵抗相关的炎性因子

摘要

目的:采用抗体芯片筛选鼻咽癌放射治疗(放疗)抵抗相关的炎性因子。方法:以放疗抵抗鼻咽癌CNE2-IR细胞与放疗敏感鼻咽癌CNE2细胞的细胞蛋白质及其细胞培养上清为样本,采用抗体芯片(AAH-INF-3)比较炎性因子的表达差异,采用免疫组织化学方法检测差异炎性因子IL-8在放疗抵抗与敏感鼻咽癌组织(每组30例)中的表达。结果:炎性因子IL-8、粒细胞-巨噬细胞集落刺激(granulocyte-macrophage colony stimulating factor,GM-CSF)和细胞间黏附分子-1(intercellular adhesion molecule 1,ICAM-1)在CNE2-IR细胞中的表达水平高于CNE2细胞,CNE2-IR细胞分泌的IL-8和GM-CSF水平也高于CNE2细胞,而CNE2-IR细胞分泌的基质金属蛋白酶-2(matrixmetalloproteinase 2,TIMP-2)水平低于CNE2细胞; IL-8在放疗抵抗鼻咽癌组织中的表达明显高于放疗敏感鼻咽癌组织。结论:IL-8,GM-CSF,ICAM-1和TIMP-2表达或分泌异常可能在鼻咽癌放疗抵抗中具有重要作用,为进一步研究鼻咽癌放疗抵抗的分子机制提供了新线索。

Identification of inflammation factors related to radioresistance of nasopharyngeal carcinoma by an antibody array

Abstract

Objective: To screen inflammation factors related to radioresistance of nasopharyngeal carcinoma (NPC).Methods: Antibody array of human inflammation factors (AAH-INF-3) were used to compare the expression differences between radioresistant NPC CNE2-IR and radiosensitive NPC CNE2 cells. Immunohistochemistry was performed to detect the expression levels of differential inflammation factor IL-8 in the radioresistant and radiosensitive NPC tissues (30 cases for each group). Results: The expression levels of IL-8, granulocyte-macrophage colony stimulating factor (GM-CSF) and intercellular adhesion molecule 1 (ICAM-1) in CNE2-IR cells were significantly higher than those in CNE2 cells, and CNE2-IR cells secreted more IL-8 and GM-CSF than CNE2 cells while secreted lesser matrixmetalloproteinase 2 (TIMP-2) than that in CNE2 cells. Immunohistochemistry showed that the expression levels of IL-8 were significantly higher in the radioresistant NPC tissues than those in the radiosensitive NPC tissues. Conclusion: Abnormal expression or secretion of IL-8, GM-CSF, ICAM-1, and TIMP-2 may play an important role in NPC radioresistance, which may provide a novel clue for further investigating the molecular mechanism of NPC radioresistance.
