姜黄素诱导黑素瘤细胞凋亡的研究

摘要

目的:研究姜黄素诱导黑素瘤细胞A375、M14凋亡的机制。方法:将对数生长的黑素瘤细胞A375、M14分别用姜黄素干预;显微镜下观察黑素瘤细胞株A375和M14的凋亡状态;应用Western Blot方法检测凋亡相关蛋白Bcl-2、Bax、Caspase-3、γ-H2AX、NF-κB的变化;应用流式细胞仪检测细胞凋亡率。结果:实验发现姜黄素干预黑素瘤细胞后可见细胞发生凋亡;在姜黄素诱导黑素瘤细胞凋亡过程中,凋亡相关蛋白Bax和γ-H2AX表达水平较对照组升高,而Bcl-2、NF-κB表达水平较较对照组下降,有活性的Caspase-3生成;随着姜黄素的浓度增加,黑色素瘤细胞早期凋亡的比例增加。结论:姜黄素诱导黑素瘤细胞生长凋亡的机制可能与其对凋亡相关蛋白Bcl-2、Bax、Caspase-3、γ-H2AX、NF-κB的表达调控有关。

Curcumin results melanoma cells apoptosis

Abstract

Objective: To observe the effects of curcumin on the apoptosis of two melanoma cell lines A375 and M14. Methods: Cell apoptosis in human melanoma cell lines A375 and M14 treated with curcumin was measured by morphological method. Furthermore four apoptosis related proteins: Bcl-2, Bax, Caspase-3, γ-H2AX and NF- κB were determined by Western Blotting analysis. Apoptosis rate were detected through FCM (flow cytometry). Results: The data showed that curcumin can induce tumor cell apoptosis. Western blotting analysis confirmed that curcumin could express increasing proteins Bax and γ-H2AX, down-regulating the proteins Bcl-2, and NF-κB, as well as, activation of Caspase-3. It showed that with the increase of the concentration of curcumin, melanoma cell early apoptosis ratio increased. Conclusion: This study showed that curcumin exhibited a remarkably increased apoptosis-inducing effects in melanoma cells.
