摘要

生物膜磷脂的不对称分布对于保证细胞代谢功能的正常进行至关重要,而其不对称性的产生和维持受P4型ATP酶(P4-ATPase)的调控。P4-ATPase是一类主动转运生物膜磷脂的蛋白,也被称为磷脂翻转酶,可以调控磷脂从细胞膜外侧或细胞器管腔面翻转到胞质面。P4-ATPase与很多生物学功能密切相关,包括囊泡运输,细胞凋亡,红细胞成熟和血液凝固等。近期研究证实P4-ATPase突变或表达异常与很多疾病的发生有关,如智力障碍、阿尔茨海默病、胆汁淤积症等。深入研究P4-ATPase的生物学功能及其与疾病的关系,有助于揭示人类相关疾病的潜在治疗靶点。

Abstract

The asymmetric distribution of phospholipids in biofilms is essential for ensuring the normal function of cell metabolism, and its construction and maintenance is regulated by P4-ATPase. P4-ATPase, also known as phospholipid flippases, is a class of proteins that can actively transport or flip phospholipids from the exocytoplasmic (extracellular or lumen) to the cytoplasmic leaflet of cellular membrane. It is essential for a wide variety of many biology processes, including vesicle transport, apoptosis, erythrocyte maturation, blood coagulation etc. Recently, study has confirmed that mutation or abnormal expression of P4-ATPase is closely associated with the pathogenesis of many diseases, such as mental retardation, Alzheimer disease, cholestasis and so forth. Further study of the biological function of P4-ATPase and its relationship with diseases will help us to find out the potential targets of human for treatment of these diseases.
