摘要

目的:探讨CXCL12、CXCR4、PI3K和AKT的表达与其在口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)淋巴结转移中的作用及各因子的相关性,以期为临床预后判断提供一定的理论基础。方法:收集2012至2015年内蒙古医科大学附属医院病理科存档的病理资料完整的口腔鳞癌手术切除组织96例,其中17例发生颈淋巴结转移为转移组,79例无颈淋巴结转移为非转移组。采用免疫组织化学法检测转移组和非转移组中CXCL12、CXCR4、PI3K和AKT蛋白的表达,分析其蛋白表达差异。结果:CXCL12、CXCR4、PI3K和AKT蛋白的表达与患者的性别、年龄、吸烟情况、分化程度和T分期均无明显相关(P>0.05),而与淋巴结转移有关(P<0.05);CXCR4与CXCL12、PI3K以及PI3K与AKT蛋白阳性表达之间均呈显著正相关(P<0.01)。结论:CXCL12、CXCR4、PI3K、AKT可能在口腔鳞癌淋巴结转移过程中相互协同,共同调控其发生。

Abstract

Objective: To explore the expression of CXCL12, CXCR4, PI3K and AKT, their role in lymph node metastasis of oral squamous cell carcinoma (OSCC) and the correlation of their factors, so as to provide a theoretical basis for clinical prognosis. Methods: We collected surgical resection tissues of OSCC archived in the Department of Pathology, Affiliated Hospital of Inner Mongolia Medical University from 2012 to 2015. Among totally 96 cases, 17 cases with cervical lymph node metastasis were grouped as the metastasis group and 79 cases without cervical lymph node metastasis were grouped into the non-metastasis group. The protein expression of CXCL12, CXCR4, PI3K and AKT in the metastatic group and the non-metastatic group were detected by immunohistochemistry, and the differences of protein expression were analyzed. Results: The protein expression of CXCL12, CXCR4, PI3K and AKT showed no significant association with gender, age, smoking status, differentiation degree, and T-staging (P>0.05), but had significant difference with lymph node metastasis (P<0.05). There was a significant positive correlation between the positive expression of CXCR4 and CXCL12, as well as PI3K and AKT (P<0.01). Conclusion: CXCL12, CXCR4, PI3K and AKT may cooperate with each other in the process of and jointly regulate the occurrence of the lymph node metastasis of OSCC.
