TRPM7在胃癌组织的表达及其临床意义

摘要

目的:探讨瞬时受体电位M型-7(transient receptor potential melastatin 7,TRPM7)在胃癌组织中的表达及临床意义。方法:收集福建省肿瘤医院2012年6月至2015年12月间进行外科手术并有完整随访信息的胃癌病例48例,应用实时荧光定量PCR验证上述病例癌组织和癌旁组织中TRPM7的表达,并分析其与临床病理的关系;应用Kaplan-Meier法行生存分析,采用Log-rank检验比较生存率。结果:TRPM7 mRNA在胃正常组织和胃癌组织中都有表达;与癌旁胃正常组织相比,TRPM7在肿瘤组织的表达比癌旁组织上调1.38倍(P=0.0140);TRPM7的表达水平与患者性别、年龄、肿瘤大小和组织分型、TNM分期及是否感染EB病毒无显著的相关性;生存分析提示:与癌旁组织相比,TRPM7正表达的胃癌患者生存时间显著短于负表达组(P=0.0442)。结论:TRPM7可能作为一个原癌基因,参与胃癌的发生和发展;TRPM7表达情况有明显的预后意义。

Abstract

Objective: To investigate the expression and clinical significance of TRPM7 in gastric cancer. Methods: A total 48 cases of gastric cancer undergoing surgery in Fujian cancer hospital from June 2012 to December 2015 were collected. The expression of TRPM7 in above cases and adjacent tissues was verified by real-time fluorescence quantitative PCR. The association between TRPM7 expression and clinicopathological features was analyzed by χ2 test. Kaplan-Meier analysis and log-rank test were used for the survival analysis. Results: TRPM7 mRNA was expressed in both normal and gastric cancer tissues. The expression of TRPM7 in tumor tissues was up-regulated by 1.38 (P=0.0140) compared with normal tissues. There was no significant correlation between the expression of TRPM7 and patients’ gender, age, size, tissue type, TNM and Epstein-Barr virus infection. Survival analysis indicated that, the survival time of gastric cancer patients with positive expression of TRPM7 was significantly lower than the group with negative expression. Conclusion: TRPM7 may be involved in the development of gastric cancer as a proto-oncogene. TRPM7 has prognostic significance in gastric cancer.
