环状非编码RNAcirc-ZNF652对卵巢癌增殖和迁移的影响及机制

摘要

目的:探讨环状非编码RNA circ-ZNF652在卵巢癌细胞系中的表达及对增殖和迁移的影响和机制。方法:采用qPCR法检测circ-ZNF652在卵巢癌细胞系PA-1,Caov-3,Caov-4,SK-OV-3及正常卵巢上皮细胞系HPOSEC,SV40中的表达。将SK-OV-3细胞分别转染circ-ZNF652敲降质粒(si-circ-ZNF652组)与阴性对照质粒(si-control组),野生型SK-OV-3细胞作为空白对照组(Control组),分别使用 CCK-8 法、细胞划痕实验检测3组细胞的细胞活力及迁移能力,蛋白质印迹法测定蛋白激酶B(Akt)和糖原合成酶激酶-3β(GSK-3β)磷酸化情况。结果:卵巢癌细胞系中circ-ZNF652表达量均显著高于正常卵巢细胞系(P<0.01);与Control组比较,si-circ-ZNF652组细胞活力降低(P<0.01)、细胞迁移能力显著降低(P<0.01),Akt和GSK-3β蛋白表达不变(P>0.05),而磷酸化Akt和磷酸化GSK-3β蛋白显著降低(P<0.01)。si-control组与Control组相比差异均无统计学意义(P>0.05)。结论:环状非编码RNA circ-ZNF652在卵巢癌细胞系中高表达,沉默其表达可抑制细胞增殖和迁移,其机制可能与调控Akt/GSK-3β信号通路有关。

Effect and mechanism of non-coding RNA circ-ZNF652 on proliferation and migration of ovarian cancer cells

Abstract

Objective: To investigate the expression of non-coding RNA circ-ZNF652 in ovarian cancer cells, its effect on proliferation and migration of ovarian cancer cells, and the potential mechanism. Methods: qPCR was used to detect the expression of non-coding RNA circ-ZNF652 in ovarian cancer cell lines PA-1, Caov-3, Caov-4, SK-OV-3 and normal ovarian cell line HPOSEC and SV40. SK-OV-3 cells were transfected with circ-ZNF652 knockdown plasmid (the si-circ-ZNF652 group) and negative control plasmid (the si-control group), whereas wild type SK-OV-3 cells were used as blank control group (the control group). CCK-8 assay and wound healing assay were used to detect the cell viability and migration ability. The expression of phosphorylation of Akt and GSK-3βwas measured by western blot. Results: The expressions of circ-ZNF652 in the ovarian cancer cell lines were significantly higher than that in the normal ovarian cell line (P<0.05). Compared with the negative control group, cells in the si-circ-ZNF652 group cells have a lower cell viability (P<0.01), cell migration was significantly decreased (P<0.01), and phosphorylationof Akt and GSK-3β were significantly suppressed (P<0.01). There was no significant difference between the si-control group and the control group (P>0.05). Conclusion: The expression of circ-ZNF652 is increased in ovarian cancer cells. The increase of its expression enhances the proliferation and migration of ovarian cancer cells, and may associate with the regulation of Akt/GSK-3β signaling pathway.
