摘要

目的:研究miR-325-3p抑制肝癌细胞增殖和转移的作用以及其作用靶点。方法:通过qRT-PCR方法研究肝癌组织以及肝癌细胞中miR-325-3p的表达情况。肝癌细胞HepG2转染miR-325-3p mimic后,通过CCK-8和Transwell法研究miR-325-3p对肝癌细胞HepG2增殖和转移的影响。通过双荧光素实验研究miR-325-3p的作用靶点。通过CCK-8和Transwell实验验证miR-325-3p与PBOV1相互作用调控HepG2细胞增殖与迁移。结果:肝癌组织和肝癌细胞中miR-325-3p表达量明显低于癌旁正常组织或正常肝细胞。miR-325-3p-mimic可显著抑制HepG2细胞增殖、侵袭和迁移。PBOV1是miR-325-3p的靶基因。与转染miR-325-3p-mimic+Lenti-Con比较,转染miR-325-3p-mimic+Lenti-PBOV1的 HepG2细胞增殖、迁移与侵袭增强。结论:MiR-325-3p通过靶向结合PBOV1来抑制肝癌细胞增殖和转移。

Abstract

Objective: To study the inhibitory effect of miR-325-3p on proliferation and metastasis of hepatocellular carcinoma cells and its target gene. Methods: The expressions of miR-325-3p in hepatocellular carcinoma tissues and hepatocellular carcinoma cells were studied by RT-qPCR. After transfection with miR-325-3p mimic into hepatocellular carcinoma cells HepG2, the effects of miR-325-3p on the proliferation and metastasis of hepatocellular carcinoma were studied by CCK-8 and Transwell assays. The target gene of miR-325-3p was studied by double fluorescein assay. The interaction between miR-325-3p and PBOV1 was proved to regulate the proliferation and migration of HepG2 cells by CCK-8 and Transwell assays. Results: The expressions of miR-325-3p in hepatocellular carcinoma tissues and hepatocellular carcinoma cells were significantly lower than that in adjacent normal tissues or normal hepatocytes. MiR-325-3p-mimic could significantly inhibit cell proliferation, invasion and migration of HepG2 cells. PBOV1 was a target gene of miR-325-3p. Compared with transfection with miR-325-3p-mimic+Lenti-Con, the proliferation, invasion and migration of HepG2 cells were increased in transfection with miR-325-3p-mimic+Lenti-PBOV1. Conclusion: MiR-325-3p inhibits proliferation and metastasis of hepatocellular carcinoma cells by targeting PBOV1.
