摘要

目的:探究妊娠期糖尿病(gestational diabetes mellitus,GDM)的孕妇体内热休克蛋白70(heat shock protein 70,HSP70)的水平及其对胎盘间质纤维化的影响。方法:检测GDM孕妇外周血和胎盘组织中HSP70水平,培养HTR-8/SVneo细胞并用高浓度葡萄糖刺激,检测HSP70被沉默前后胶原蛋白I(collagen I,COL I)、COL IV和FN1的表达,Western印迹法检测Smad7的磷酸化水平。结果:GDM孕妇体内的HSP70水平显著高于正常孕妇(P<0.01),高糖刺激下HTR-8/SVneo细胞中COL I,COL IV和FN1的表达上升(P<0.01),Smad7的磷酸化水平上调(P<0.01)。当转染HSP70 siRNA后COL I,COL IV和纤维粘连蛋白1(fibronectin 1,FN1)的表达下及Smad7的磷酸化水平均下调。 结论:GDM孕妇体内HSP70水上升,并通过Smad7通路促进胎盘组织纤维化。

Serum HSP70 levels increases in pregnant women with gestational diabetes and promotes placental interstitial fibrosis

Abstract

Objective: To determine the heat shock protein 70 (HSP70) level in pregnant women with gestational diabetes mellitus and its role in placenta mesenchymal fibrosis. Methods: HSP70 level in peripheral blood and placenta were determined. HTR-8/SVneo cells were cultured and treated with high concentration glucose. The levels of COL I, COL IV and FN1 were detected before and after HSP70 silencing. The phosphorylation of Smad7 was measured by Western blot. Results: HSP70 levels in pregnant women with gestational diabetes mellitus were significantly higher than those without gestational diabetes mellitus (P<0.01). The levels of COL I, COL IV and FN1 in HTR-8/SVneo cells were significantly up-regulated when treated with high concentration of glucose and were down-regulated after HSP70 silencing (P<0.01). The phosphorylation of Smad7 were increased when treated with high concentration of glucose and were decreased after HSP70 silencing (P<0.01). Conclusion: Increase of HSP70 levels induce placenta mesenchymal fibrosis via the Smad7 pathway in pregnant women with gestational diabetes mellitus.
