摘要

目的:探讨黄连素对多发性骨髓瘤细胞活性和凋亡的影响。方法:通过CCK-8试验检测不同浓度黄连素作用下多发性骨髓瘤U266细胞的增殖;ELISA试验检测不同浓度黄连素作用下U266分泌IL-6的情况;细胞凋亡试验检测不同浓度黄连素作用下U266骨髓瘤细胞凋亡行为变化;透射电子显微镜(transmission electron microscope,TEM)和扫描电子显微镜(scanning electron microscope,SEM)观察U266细胞的超微结构。结果:随着浓度(40~160 μmol/L)的改变,黄连素以剂量和时间依赖的方式抑制U266细胞细胞增殖和IL-6分泌;黄连素以剂量依赖的方式诱导U266细胞G2/M期停滞和凋亡,80 μmol/L黄连素处理后的U266细胞的超微结构发生凋亡的形态特征。结论:黄连素能抑制人多发性骨髓瘤细胞的活性,诱导骨髓瘤细胞G2/M期停滞并诱导细胞凋亡。

Effects of berberine on proliferation and apoptosis of multiple myeloma cells

Abstract

Objective: To investigate the effect of berberine on the activity and apoptosis of multiple myeloma cells. Methods: The proliferation of multiple myeloma U266 cells under different concentrations of berberine were detected by CCK-8. ELISA was used to detect the secretion of IL-6 by U266 under different concentrations of berberine. The apoptosis of U266 myeloma cells was detected by flow cytometry under different concentrations of berberine. The ultrastructure was observed by transmission electron microscopy (TEM) and scanning electron microscopy (SEM). Results: Treatment of U266 cells with berberine (40–160 µmol/L) suppressed cell proliferation and IL-6 secretion in dose- and time-dependent manners. At the same time, berberine dose-dependently induced G2/M phase arrest and apoptosis in U266 cells. The ultrastructure of U266 cells treated with 80 μmol/L berberine was morphologically characterized by apoptosis. Conclusion: Berberine can inhibit the activity of human multiple myeloma cells, induce G2/M stagnation and induce apoptosis.
