柴芪汤对非酒精性脂肪肝大鼠模型血清TNF-α和IL-6表达的影响

摘要

目的:观察柴芪汤对非酒精性脂肪肝(non-alcoholic fatty liver disease,NAFLD)大鼠炎症因子TNF-α和IL-6的影响,探讨其延缓NAFLD进展的作用机制。方法:SPF级雄性SD大鼠40只,随机分为空白组、模型组和治疗组,高脂饲料喂养造模,8周后,空白组与模型组各取8只处死取材,评价模型。造模成功后治疗组每天给予柴芪汤5.67 g/kg灌胃,8周后,处死各组动物,检测血清AST,ALT,TG,TC,LDL-C,HDL-C,TNF-α及IL-6水平,空腹血糖(fasting blood glucose,FBG)及胰岛素(fasting insulin,FINS)水平,评价胰岛素抵抗指数(insulin resistance index,IRI);取肝组织,镜下观察病理形态。结果:高脂饲养8周后,模型组大鼠血清ALT,AST,FBG,FINS及IRI水平高于空白组,差异有统计学意义(P<0.05),可见肝细胞脂肪变性;柴芪汤治疗8周后,大鼠血清ALT,AST,TG,TC,LDL-C,TNF-α及IL-6水平低于模型组,差异有统计学意义(P<0.05),肝细胞脂肪变性程度较模型组轻,肝细胞坏死及炎性细胞浸润不明显。结论:高脂饲养8周后,NAFLD大鼠模型制备成功;柴芪汤可减轻NAFLD大鼠模型肝脏炎症反应和肝细胞脂肪变性程度,下调炎性因子TNF-α和IL-6的表达,降低血清TG,TC及LDL-C,改善胰岛素抵抗。

Effect of Chaiqi decoction on the level of TNF-α and IL-6 in mouse model with non-alcoholic fatty liver disease

Abstract

Objective: To observe the influence of Chaiqi decoction on the level of TNF-α and IL-6 in serum of mice model with NAFLD, and study the mechanism of dispersing stagnated liver and invigorating spleen therapy in preventing the progress of NAFLD. Methods: Forty SD mice were randomized into a normal group, a NAFLD model group and a Chaiqi decoction treat group. The mice, except the normal group, were fed with high fat for 8 weeks. Then 8 mice in the normal group and the NAFLD model group were executed respectively to evaluate the model of NAFLD. After the model made, the treat group accepted gastric lavage of Chaiqi decoction (5.67 g/kg) each day for 8 weeks. Then all mice were executed to detect the level of TG, TC, LDL-C, HDL-C, TNF-α and IL-6 in serum of mice with NAFLD, and the insulin resistance index (IRI) from fasting blood glucose (FBG) and fasting insulin (FINS). The pathology of liver were observed through microscope. Results: After fed with high fat diet for 8 weeks, the level of ALT, AST, FBG, FINS and IRI in serum of mice were higher than that in the normal group, and the differences were statistically significant (P<0.05), fatty degeneration can be seen in liver cells. After threated with Chaiqi decoction for 8 weeks, the level of ALT, AST, TG, TC, LDL-C, TNF-α and IL-6 in serum were lower than that in the model group, the differences were statistically significant (P<0.05), fatty degeneration in liver cells were better than the mice in model group. Conclusions: After fed with high fat diet for 8 weeks, we succeed in building up NAFLD model in mice. Chaiqi decoction can reduce the level of inflammatory response and fatty degeneration in liver cells, lower down the expression of TNF-α and IL-6 in serum. It also can lower the level of TG, TC and LDL-C in in serum of mice with NAFLD and improve IR, too.
