摘要

目的:研究p62在鼻咽癌(nasopharyngeal carcinoma,NPC)细胞、组织中的表达情况及其自身抗体在外周血中的水平,探究p62与NPC发生、转移的关系及其在临床中的应用价值。方法:采用Western印迹法和免疫组织化学染色法分别检测NPC细胞株NP69,6-10B和5-8F和NPC患者组织中的p62表达;ELISA检测患者血清中抗p62抗体浓度,并分析其与临床病理特征的关系。结果:p62在NP69,6-10B和5-8F细胞中的表达水平依次升高(P<0.05);NPC组织中p62高表达,其水平与患者有无转移(χ2=9.332,P=0.002)有关。NPC患者血清中抗p62水平较慢性鼻咽炎组明显升高(t=–4.653,P<0.001),且抗p62抗体水平与患者是否转移有关(t=14.255,P=0.016);血清抗p62抗体筛查NPC的最佳临界值为51.82 μg/L,判别是否发生转移的临界值为62.03 μg/L。结论:p62高表达与NPC发生及转移密切相关,组织中p62蛋白和血清中抗p62抗体可能作为NPC病情监测和判断预后的新指标。

Expression of p62/SQSTM1 in nasopharyngeal carcinoma

Abstract

Objective: To study the expression of p62 in nasopharyngeal carcinoma (NPC) cell lines and tissues, and explore the serum concentration of anti-p62 in NPC patients, and investigate the correlation between p62 and NPC occurrence and metastasis, and assess its clinical value. Methods: The expression of protein p62 in NPC cell lines (NP69, 6-10B and 5-8F) was detected by Western blot. Immunohistochemistry staining method was used to test the expression of p62 in NPC tissue specimens. Serum anti-p62 antibody level was determined by enzyme-linked immunosorbent assay, then analyzing its relationship with clinical pathological features. Results: The expression of p62 in NP69, 6-10B and 5-8F cells was increased successively (P<0.05); the expression of p62 was increased in nasopharyngeal carcinoma tissues, which was closely correlated with metastasis (χ2=9.332, P=0.002). Compared with chronic rhinopharyngitis group, the serum level of anti-p62 in NPC patients was dramatically increased (t=–4.653, P<0.001) and correlated with metastasis (t=14.255, P=0.016); the optimal value of serum anti-p62 screening for NPC is 51.82 μg/L, and the critical value of anti-p62 judging NPC patients whether metastasis or not is 62.03 μg/L. Conclusion: High expression of p62 is closely related to the occurrence and metastasis of NPC, p62 protein in tissues and serum anti-p62 may be used as new indicators for monitoring the conditions and prognosis of NPC patients.
